摘要
目的:建立大鼠组织中橙皮苷与川陈皮素的浓度测定方法,考察橙皮苷和川陈皮素在大鼠体内的组织分布,探究不同组织作用趋势。方法:大鼠单次灌胃橙皮苷50 mg/kg和川陈皮素25 mg/kg后,分别于0.25、0.5、1、1.5、2、4、6、12、24 h采集血浆、心、肝、脾、肺、肾、脑、肌肉、小肠、大肠、胃、脂肪等各组织样品,采用UPLC-MS检测橙皮苷与川陈皮素的组织匀浆浓度。结果:建立的UPLC-MS测定方法简便快速,专属性、基质效应、准确度、回收率和稳定性良好。橙皮苷在大鼠的小肠和胃中浓度最高,其次为大肠、肌肉、脂肪;川陈皮素在大鼠的小肠和肝组织中分布浓度最高,其次是胃、脂肪、肾和大肠。结论:橙皮苷和川陈皮素在大鼠体内分布均较广泛,该研究结果为陈皮中黄酮类成分的进一步研究开发提供参考。
Objective:To establish a method for the concentration determination of hesperidin and nobiletin in rat tissues,and to investigate the tissue distribution in rats.Methods:After a single oral administration of hesperidin(50 mg/kg)and nobiletin(25 mg/kg),samples of plasma,heart,liver,spleen,lung,kidney,brain,muscle,small intestine,large intestine,stomach,and fat were collected at 0.25,0.5,1,1.5,2,4,6,12,and 24 hours,respectively.The homogenate concentrations of hesperidin and nobiletin in rat tissues were detected by UPLC-MS.Results:The UPLC-MS method was simple and rapid,with good specificity,matrix effect,accuracy,recovery rate,and stability.Hesperidin was widely distributed,and its highest concentration was found in small intestine and stomach,followed by large intestine,muscle,and fat.Nobiletin was distributed in all tissues,and the highest concentration was found in small intestine and liver tissues,followed by stomach,fat,kidney,and large intestine.Conclusion:Hesperidin and nobiletin are widely distributed in rats.The findings of this study provide references for the further research and development of flavonoids in CITRI RETICULATAE PERICARPIUM.
