非水毛细管电泳法测定葛根中葛根素和大豆苷元的含量

摘要 目的:建立非水毛细管电泳法快速分离测定具有神经功效的两种中药活性成份(葛根素和大豆苷元)并应用于葛根中两种组分的含量测定。方法:采用非水毛细管电泳法,以七叶内酯作为内标,90 mmol·L-1胆酸钠-3.0%乙酸-15%乙腈的甲醇液作为缓冲液,熔融石英毛细管柱分离电压:20 kV,检测波长:254 nm,柱温:25℃压力进样:50 mbar×5 s。结果:葛根素和大豆苷元在10~200μg·ml-1与峰面积线性关系良好(r=0.9991和r=0.9990),平均加样回收率分别为96.72%和97.26%,RSD分别为1.94%和2.17%(n=5)。结论:该方法简单、快速、准确、重复性好,可用于葛根的两种组分的含量测定。 Objective:To establish a non-aqueous capillary electrophoresis(NCAE)method for the determination of puerarin and daidzein in Puerariae Radix.Methods:The separation was carried out on an uncoated fused capillary with a running buffer containing90 mmol·L-1 sodium cholate,3.0%acetic acid and 15%acetonitrile in methanol.Esculetin was used as the internal standard.A separation voltage of 20 kV and the UV detector wavelength at 254 nm were adopted.The column temperature was 25℃and the pressure injection was 50 mbarx5 sec.Results:The calibration ranges were 10-20μg·ml-1 for both analytes(r=0.9991 and r=0.9990).The average recovery was 96.72%(RSD=1.94%,n=5)for puerarin and 97.26%(RSD=2.11%,n=5)for daidzein in Puerariae Radix.Conclusion:The proposed method is simple,rapid,accurate and highly repeatable,and can be used to control the quality of Puerariae Radix.
