摘要
目的:研究海金沙地上部分的化学成分及对银屑病角质形成细胞的影响。方法:海金沙地上部分90%乙醇提取物采用硅胶、Sephadex LH-20葡聚糖凝胶分离纯化,根据理化性质及波谱数据鉴定化合物的结构。采用MTT法测试化合物对银屑病角质形成细胞的影响。结果:从海金沙地上部分中分离得到13个化合物,分别鉴定为:japonica acid(1)、5,3′,4′-三羟基-7-甲氧基二氢黄酮(2)、burselignan(3)、acetylwikstresinol(4)、5,6,7-三甲氧基香豆素(5)、dihydrodehydrodiconiferyl alcohol(6)、甘草黄烷酮(7)、prismaconnatoside(8)、northalifoline(9)、2,3-二羟基-1-(4-羟基-3,5-二甲氧基苯基)-1-丙酮(10)、2,4,6-trichlorol-3-methyl-5-methoxy-phenol-1-O-β-D-glucopy-ranosyl-(1→6)-β-D-glucopyranoside(11)、licorice-saponin G2(12)、对羟基苯乙酮(13)。化合物1、7、9、11、12对银屑病角质形成细胞增殖具有不同程度的抑制作用。结论:所有化合物均为首次从海金沙中分离得到,具有一定的抗银屑病角质形成细胞增殖的作用。
Objective:To study the chemical constituents of Lygodium japonicum aboveground part and the effects on psoriatic keratinocytes.Methods:The 90%ethanol extracts of Lygodium japonicum aboveground part were isolated and purified by silica gel and Sephadex LH-20.The structures of the compounds were identified by physicochemical properties and spectral data.The effect of compounds on psoriatic keratinocytes was tested by MTT assay.Results:Thirteen compounds were isolated from Lygodium japonicum aboveground part and identified as japonica acid(1),5,3′,4′-trihydroxy-7-methoxyflavanone(2),burselignan(3),acetylwikstresinol(4),5,6,7-trimethoxy coumarin(5),dihydrodehydrodiconiferyl alcohol(6),licoflavanone(7),prismaconnatoside(8),northalifoline(9),2,3-dihydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-1-propanone(10),2,4,6-trichlorol-3-methyl-5-methoxy-phenol-1-O-β-D-glucopy-ranosyl-(1→6)-β-D-glucopyranoside(11),licorice-saponin G2(12),p-hydroxyacetophenone(13),respectively.Compounds 1,7,9,11 and 12 showed different degrees of inhibitory effects on keratinocytes proliferation in psoriasis.Conclusion:All compounds are isolated from Lygodium japonicum for the first time,Lygodium japonicum has a certain anti-psoriatic keratinocytes.
