摘要
目的:建立骨碎补生品、砂烫品、微波炮制品的指纹图谱及4个成分的含量测定方法,并结合多元统计分析方法研究差异性。方法:采用HPLC法测定骨碎补生品及炮制品的指纹图谱及5-羟甲基糠醛、原儿茶酸、新北美圣草苷、柚皮苷4个成分的含量,结合相似度评价、单因素方差分析、主成分分析、聚类分析、正交偏最小二乘判别分析法对指纹图谱进行差异性分析。结果:骨碎补经砂烫、微波炮制后新增5-羟甲基糠醛,原儿茶酸的平均含量增加;新北美圣草苷、柚皮苷的平均含量降低。骨碎补生品、砂烫品、微波炮制品指纹图谱分别标定了11、14、15个共有峰,相似度均>0.890。多元统计分析均可明显区分生品与炮制品,并发现5个区分生品与炮制品的标志性色谱峰。结论:该研究建立的方法可有效区分骨碎补生品与炮制品,为骨碎补生品与炮制品的鉴别及质量分析提供参考。
Objective: To establish a method for the fingerprints and content determination of four components of Drynariae Rhizoma raw products, sand-scorch products, microwave products, and to study the differences with multivariate statistical analysis.Methods: HPLC method was used to determine the fingerprints and the contents of 5-hydroxymethylfurfural, protocatechuic acid, neoeriocitrin and naringin in Drynariae Rhizoma raw products and it different processed products.The similarity evaluation, one-way analysis of variance, principal component analysis, cluster analysis and orthogonal partial least squares discriminant analysis were used to analyze the differences of the fingerprints.Results: 5-Hydroxymethylfurfural was added after sand-scorch and microwave processing.The average content of protocatechuic acid increased, the contents average of neoeriocitrin and naringin decreased.11,14 and 15 common peaks were calibrated for the fingerprints of Drynariae Rhizoma raw, sand-scorch and microwave processed products, respectively, and the similarity degree was greater than 0.890.Multivariate statistical analysis could clearly distinguish raw and processed products, and five symbolic chromatographic peaks were found to distinguish raw and processed products.Conclusion: The method established in this study can effectively distinguish the raw and processed products of Drynariae Rhizoma, and provide reference for the identification and quality analysis of raw and different processed products of Drynariae Rhizoma.
