摘要
目的:运用网络药理学技术,探寻紫花地丁抗痛风/高尿酸血症的作用机制。方法:以紫花地丁为关键词,在TCMSP、MESH数据库检索其已知成分;在TCMSP、Drugbank和SwissTargetPrediction检索紫花地丁已知成分的靶点基因;同时,以痛风/高尿酸血症为关键词,在TTD、Drugbank、DisGeNET数据库检索相应疾病的靶点基因;运用ImageGP平台将成分靶点和疾病靶点映射,得到紫花地丁作用于痛风/高尿酸血症的预测靶点;运用Cytoscape3.2.1软件的ClueGO插件和DAVID数据库进行GO生物学过程(GO-BP)分析和KEGG富集分析,并构建“化合物-靶点-疾病-通路”网络。结果:得到紫花地丁有效成分27个,相关靶点151个;疾病相关靶点226个;并通过ImageGP平台获得紫花地丁和疾病的共同靶点24个;共同靶点主要富集于12条GO-BP和10条KEGG信号通路上发挥治疗痛风/高尿酸血症的作用。结论:本研究初步探讨了紫花地丁抗痛风/高尿酸血症的作用机制,为进一步研究奠定了基础。
Objective: To investigate the mechanism of Violae Herba on anti-gout/hyperuricemia based on network pharmacology.Methods: The chemical constituents were collected based on the TCMSP and MESH databases by using Violae Herba as the key word;The target genes of known compositions in Violae Herba were predicted by TCMSP,Drugbank and SwissTargetPrediction databases;At the same time, the target genes for corresponding diseases were searched by TTD,Drugbank and DisGeNET databases using gout and hyperuricemia as the key words;The component targets and disease targets were mapped by the ImageGP platform to obtain the predicted targets of Violae Herba acting on gout/hyperuricemia.The ClueGO plug-in of Cytoscape3.2.1 software and DAVID database were used for GO biological process(GO-BP)analysis and KEGG enrichment analysis, and the“compounds-targets-diseases-pathways”network was constructed.Results: Twenty-seven active components of Violae Herba and 151 related targets were obtained.226 disease-related targets were obtained;Through the ImageGP platform, 24 common targets of Violae Herba and disease were obtained.The common targets were mainly concentrated in 12 GO-BP and 10 KEGG signaling pathways to play a role in the treatment of gout/hyperuricemia.Conclusion: In this study, the mechanism of Violae Herba on anti-gout/hyperuricemia is preliminarily investigated, which lays a foundation for further research.
