金线莲乙醇提取物对自然衰老小鼠抗衰老作用的研究 被引量:6

摘要 目的:研究金线莲乙醇提取物对自然衰老小鼠的抗衰老作用。方法:选取60只12月龄昆明小鼠,按体质量随机分为模型组、维生素E阳性对照组及金线莲乙醇提取物低、中、高剂量组。各组灌胃给药,模型组给予等体积生理盐水,1次/d,连续4 w,每周测1次小鼠体质量。连续灌胃给药3 w后,利用Morris水迷宫评估小鼠空间学习与记忆能力。第4周后处死,采用HE染色法观察小鼠海马组织病理学变化;测定小鼠脑组织中MDA、SOD、GSH-Px、ACh-E、MAO水平,皮肤中CAT、HYP水平及肝组织中AGEs水平。结果:与模型组比较,给药期间小鼠体质量差异无统计学意义;金线莲乙醇提取物可显著缩短自然衰老小鼠逃避潜伏期及到达平台路程,同时改善海马神经细胞的固缩深染;显著降低脑组织MDA、ACh-E、MAO水平,并显著增强抗氧化酶(SOD、GSH-Px)活性;显著提高皮肤中CAT、HYP水平;并显著降低肝脏中AGEs水平。结论:金线莲乙醇提取物具有一定抗衰老作用,能改善自然衰老小鼠的学习与记忆能力和海马神经细胞病变,其机制可能与提高机体抗氧化酶活性及降低氧化物水平有关。 Objective:To investigate the anti-aging effect of ethanol extract from Anoectochilus roxburghii(AREE)in senescent mice.Methods:Sixty 12-month-old KM mice were randomly divided into model group,vitamin E positive control group and low-,medium-and high-dose Anoectochilus roxburghii ethanol extract groups according to body mass.Each group was given the drug by gavage,the model group was given the same volume of normal saline,1 time/d,4 w continuously,the body mass of the mice was measured once a week.After continuous administration of 3 w,Morris water maze was used to evaluate the spatial learning and memory ability of mice.The mice were sacrificed after 4 w,the histopathological changes of hippocampal tissues were observed by HE staining.The levels of MDA,SOD,GSH-Px,ACh-E and MAO in the brain tissue,CAT and HYP in the skin,AGEs in the liver tissue were measured.Results:Compared with the model group,there was no significant difference in body mass during administration.The ethanol extract from Anoectochilus roxburghii could significantly shorten the escape incubation period and reach the platform in senescent mice and improve the hyperchromatin of hippocampal nerve cells,significantly reduce the levels of MDA,ACh-E and MAO in the brain tissue,and significantly enhance the activities of antioxidant enzymes(SOD,GSH-Px),significantly increase CAT and HYP levels in the skin,significantly reduce AGEs level in the liver.Conclusion:The ethanol extract of Anoectochilus roxburghii has a certain anti-aging effect,can improve of learning and memory ability,hippocampal neurocytosis of senesent mice,its mechanism may be related to the improvement of antioxidant enzyme activity and the reduction of oxide level.
