基于欧洲人群公共数据的代谢综合征多组分共享遗传研究

Study of shared heritability in multi-components of metabolic syndrome based on public data of European population

目的研究代谢综合征各组分之间的遗传相关性,探讨不同性状共享的遗传组分对各性状贡献的大小,探索代谢综合征各组分间的共享遗传基础。方法利用欧洲人群收缩压、舒张压、空腹血糖、餐后2 h血糖、腰围、甘油三酯和高密度脂蛋白胆固醇的全基因组关联研究(GWAS)汇总统计数据以及连锁不平衡评分数据,采用R 3.5.0软件的GenomicSEM软件包构建跨性状连锁不平衡评分回归(cross-trait LDSC),计算各性状之间的遗传相关性。应用共同因子分析探讨不同性状共享的遗传组分对各性状贡献的大小。结果收缩压和舒张压具有显著正向的遗传相关性(r_g=0.81),高密度脂蛋白胆固醇与甘油三酯、腰围均具有负向的遗传相关性(r_g值分别为-0.61、-0.41),均有统计学意义(P<0.05)。共享因子分析结果显示,共享遗传因素对腰围、甘油三酯和高密度脂蛋白胆固醇这3个性状的因子载荷分别为0.456、0.678和-0.903。结论在代谢综合征多组分的相关性状中,欧洲人群收缩压与舒张压,腰围、高密度脂蛋白胆固醇与甘油三酯存在一定程度的共享遗传。

Objective To study the genetic correlation among the multi-components of metabolic syndrome,and to explore the contribution of the shared genetic components of metabolic syndrome to each trait,and to explore the shared heritability basis among the components of metabolic syndrome.Methods The genome-wide association analysis(GWAS)summary statistics data of systolic blood pressure,diastolic blood pressure,fasting blood glucose(FBG),2 h postprandial blood glucose,waist circumference(WC),triglyceride(TG)and high-density lipoprotein cholesterol(HDL-C)and the data of linkage disequilibrium(LD)score in the European population were used to calculate the genetic correlation among traits with construction of cross trait linkage disequilibrium score regression(cross-trait LDSC)in Genomic SEM of R 3.5.0 software.Common factor analysis was used to explore the contribution of the different trait shared genetic components to traits.Results There was significant positive correlation between systolic pressure and diastolic blood pressure(r_g=0.81).There was significant negative correlation between HDL-C and TG or between HDL-C and WC(r_g values were-0.61 and-0.41),P<0.05.Shared genetic factor analysis showed that the factor loadings of shared genetic factors on WC,TG and HDL-C were 0.456,0.678 and-0.903,respectively.Conclusion Among the multi-components of metabolic syndrome,the systolic blood pressure and diastolic blood pressure,WC,HDL-C and TG of European population possesses the shared heritability to a certain extent.
