摘要
为确定芽孢杆菌腐熟菌剂的高效固态发酵工艺参数并研究该菌剂在促进果树枝条堆肥进程中降解的效果,采用单因素试验结合多因素正交试验的方法对芽孢杆菌的发酵参数进行了优化,并测定了该菌剂对果树枝条堆肥过程中纤维素与木质素降解的影响。结果表明,在麸皮固体培养基中添加30%玉米粉、0.2%葡萄糖、0.1%胰蛋白胨、0.015%硫酸镁,控制培养基接种量为20%、固液比为1∶0.8、发酵温度为28℃即可实现芽孢杆菌的高效培养,有效菌落数最高达1150.00×10^(8) cfu/g。此外,果树枝条中添加2.5%芽孢杆菌腐熟菌剂进行堆肥可显著促进纤维素与木质素的降解,降解率分别提高191.00%与107.01%。
In order to determine the suitable parameters for bacillus decomposing agent in the solid-state fermentation and evaluate its effect on promoting the degradation of fruit branches in composting process. The optimum fermentation parameters of bacillus were determined by single factor test and multi-factor orthogonal test in this study, and the effect of this agent on lignin and cellulose degradation in fruit branch composting was verified. The results showed that in the solid-state fermentation of bacillus agent, the highest colony count of 1 150.00×10^(8)cfu/g was achieved in bran medium with the addition of 30% corn meal, 0.2% glucose, 0.1% tryptone and 0.015% magnesium sulfate. And the fermentation conditions were controlled at the inoculation amount of 20%, solid-liquid ratio of 1∶0.8 and fermentation temperature of 28 ℃. Furthermore, the degradation rates of cellulose and lignin increased by 191.00% and 107.01% by adding 2.5% bacillus agent to the fruit branch during the composting.
