摘要
目的:探讨三颗针炮制前后对CCI_(4)、酒精性肝损伤小鼠的保肝作用及体外抗氧化作用的影响。方法:分别采用CCI_(4)、56°白酒建立小鼠肝损伤模型,分别连续给药7d后,收集肝组织标本与小鼠血清,检测肝组织中的丙二醛(MDA)、超氧化歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)及血清中的谷草转氨酶(AST)、谷丙转氨酶(ALT)含量,计算肝脏指数。通过测定三颗针不同制品清除羟自由基及1,1-二苯基-2-三硝基肼自由基(DPPH)的能力,评价其抗氧化活性。结果:三颗针各制品可显著性抑制CCI_(4)致小鼠肝损伤血清中AST与ALT活性升高(P<0.01),可显著性降低56°白酒所致肝损伤肝组织中MDA水平(P<0.01),醋制、酒制与同剂量生品MDA水平有显著性差异(P<0.01),各制品高剂量可显著性升高SOD与GSH-Px活力。三颗针生品、醋制品、酒制品及VC体外清除羟自由基的IC_(50)值分别是3.103、2.212、2.485、0.607mg;清除DPPH自由基的IC_(50)值分别是0.073、0.042、0.049、0.029mg。结论:三颗针对CCI_(4)、56°白酒致小鼠急性肝损伤具有保护作用,并具一定体外抗氧化作用,炮制后保肝与抗氧化作用增强。
Objective:To discussing the effect of Berberis Radix for CCI_(4),hepatic protection and antioxidation in vitro of hepatic injured mice by alcohol before and after processing.Methods:To build hepatic injured models of CCI_(4)and 56%alc/vol;collecting hepatic tissue specimens and serum after seven days dosing;detecting the content of MDA,SOD and GSH-Px in hepatic tissue and AST,ALT in serum;calculating the hepatic index.Estimating the antioxidation by measuring the scavenging capacity of Berberis Radix in different products of Hydroxyl radical and 1,1-diphenyl-2-DP-PH.Results:All manufactured products of Berberis Radix can obviously restrain the increasing of AST and ALT of hepatic serum which is injured by CCI_(4)(P<0.01)and reduce the MDA of hepatic tissue which is injured by 56%alc/vol(P<0.01).There is a obvious difference of MDA by comparing the Wine-Processed,Vinegar-Processed and unprocessed models in same dosage(P<0.01).The manufactured products in high dosage can rise the vitality of SOD and GSH-Px obviously.The value of IC_(50)to scavenge Hydroxyl radical in vitro is respectively 3.103,2.212,2.485,0.607(mg)and the value of IC_(50)to scavenge DPPH is respectively 0.073,0.042,0.049,0.029(mg)in unprocessed,Vinegar-Processed,Wine-Processed and VC-processed Berberis.Conclusion:Berberis Radix has a protective effect against acute hepatic injury by CCI_(4)and 56%alc/vol,but also has some antioxidation in vitro.And it can enhance the hepatic protection and antioxidation after processing.
