正交试验优化黑果腺肋花楸果实中绿原酸提取工艺研究 被引量:4

摘要 目的:采用正交试验优化黑果腺肋花楸果实中绿原酸的提取工艺。方法:采用超声法提取黑果腺肋花楸果实中的绿原酸,运用紫外分光光度法测定其含量。以绿原酸提取率为评价指标,设计5个单因素,依据单因素考察筛选出主要因素,并采用正交试验优化绿原酸的提取工艺。结果:绿原酸的最优提取工艺为乙醇体积分数60%、料液比1∶40(g/mL)、提取时间60 min、提取温度55℃。在此提取工艺下验证3次,得到绿原酸含量分别为12.3 mg/g、12.15 mg/g、11.95 mg/g,平均含量为12.13 mg/g,提取率分别为1.207%、1.186%、1.167%,平均提取率为1.187%,RSD为0.07%。结论:正交试验法可有效提高黑果腺肋花楸果实中绿原酸的提取率,结果重复性好、稳定可行。 Objective:To Optimize the extraction process of chlorogenic acid from the fruit of the Aronia melanocarpa.Methods:The chlorogenic acid in the fruit was extracted by ultrasonic method,and its content was determined by ultraviolet spectrophotometry.Based on the evaluation index of chlorogenic acid extraction rate,five single factors were designed,and the main factors were selected according to single factor investigation.The orthogonal experiment was used to optimize the extraction process.Results:The optimal extraction process was obtained with ethanol volume fraction of 60%,solid-liquid ratio of 1:40(g/mL),extraction time of 60 min,extraction temperature of 55℃,and three measurements under the extraction process.The chlorogenic acid content was 12.3 mg/g,12.15 mg/g,11.95 mg/g,average content 12.13 mg/g,extraction rate1.207%,1.186%,1.167%,average extraction rate 1.187%,RSD 0.07%.Conclusion:The method can effectively improve the extraction rate of chlorogenic acid in the fruit of theAornia mealnocarpa.The result is good repeatability,stable and feasible,and high content.
