六神曲抗菌有效部位化学成分研究

Study on the chemical constituents of antibacterial part from Medicated Leaven Massa

目的对中药六神曲抗菌有效部位进行化学研究,以明确其药效物质基础。方法采用溶剂萃取法制备六神曲抗菌有效部位,并利用各种柱色谱方法对六神曲抗菌有效部位进行化学成分分离,经各种波谱数据和理化分析,对分离得到的化合物进行结构鉴定。结果共分离鉴定了11个化合物,包括3个生物碱类化合物:川芎哚(1)、3-羧基川芎哚(2)、(1S,3S)-1-甲基-1,2,3,4-四氢-β-咔啉-3-羧酸(3);2个含氮类化合物:尿囊素(4)和腺苷(5);2个羟脂类化合物:pinellic acid(6)和(9S,10E,12S,13S)-9,12,13-三羟基-10-十八烯酸甲酯(7);1个苯丙素类化合物:阿魏酸(8);2个芳香族化合物:它乔糖苷(9)和香草酸(10);1个甾体皂苷类化合物:β-胡萝卜苷(11)。结论上述成分均为首次从六神曲中分离得到,已有文献报道表明,部分分离得到的化合物具有显著的抗菌或抗病毒感染活性,这些成分很可能是六神曲发酵过程中产生的次生代谢产物或生物转化产物,即六神曲发挥其临床疗效的物质基础。

Objective To elucidate the therapeutic basis of Medicated Leaven Massa(MLM)by studying on the constituents of its antibacterial part.Methods The antibacterial fraction of MLM was prepared by solvent extraction,and then separated with various chromatographic columns.The structures of the obtained compounds were identified by physicochemical analysis and various spectra data.Results Eleven compounds were isolated from the antibacterial fraction of MLM,including three alkaloids of perlolyrine(1),3-carboxyl-perlolyrine(2),(1 S,3 S)-1-methyl-1,2,3,4-tetrahydro-β-carboline-3-carboxylic acid(3),two compounds with nitrogen of allantoin(4)and adenosine(5),two hydroxyl lipids of pinellic acid(6)and methyl(9 S,10 E,12 S,13 S)-9,12,13-trihydroxy-10-octadecenoate(7),a phenylpropanoid of ferulic acid(8),two aromatics of(tachioside,9)and vanillic acid(10),and a steroidal saponin of(β-daucosterol,11).Conclusion The chemical study was conducted on the antibacterial fraction of MLM for the first time.Some isolated compounds with significant antibacterial and antivirus infection had been reported in previous references.These compounds probably be the secondary metabolites or biotransformation products produced during the fermenting process of MLM,i.e.the therapeutic basis of MLM.
