摘要
目的观察2型糖尿病大鼠模型血清中氧化应激反应因子ROS、MDA、SOD及胰腺组织中PDX-1、MafA mRNA在大柴胡汤干预治疗前后的变化,探讨大柴胡汤对2型糖尿病大鼠胰岛细胞的保护机制。方法50只雄性SD大鼠随机选取10只作为空白组,剩余40只给予高糖高脂饲料喂养加腹腔注射链脲佐菌素(STZ)复制2型糖尿病大鼠模型,成模后随机分为大柴胡汤组、辛伐他汀组、二甲双胍组和模型对照组。模型对照组、正常对照组给予生理盐水10 ml·kg-1灌胃,大柴胡汤组、辛伐他汀组、二甲双胍组分别给予大柴胡汤10.1 g/kg/d、辛伐他汀混悬液20 mg/kg/d、二甲双胍混悬液300 mg/kg/d灌胃,给药8周后检测取材检测相关指标。结果与模型组比较,大柴胡汤组FBG、HbA1c、TC、TG、FFA水平降低,PDX-1、MaFA mRNA表达上升(P<0.05或P<0.01)。结论大柴胡汤治疗2型糖尿病的作用机制可能是抑制氧化应激反应而达到保护胰岛β细胞作用。
Objective To observe the changes of oxidative stress response factors ROS,MDA,SOD and PDX-1 and MafA mRNA in pancreatic tissue before and after treatment with Dachaihu Decoction in rats with type 2 diabetes mellitus.Protective mechanism of islet cells in diabetic rats.Methods Fifty male Sprague-Dawley rats were randomly selected as a blank group.The remaining 40 rats were fed with high-sugar and high-fat diet and intraperitoneal injection of streptozotocin(STZ)to replicate type 2 diabetic rat models.Da Chai Hu Tang group,simvastatin group,metformin group and model control group.The model control group and the normal control group were given normal saline 10 ml/kg,and the Dachaihutang group,simvastatin group and metformin group were given Dachaihu Decoction 10.1 g/kg/d and simvastatin suspension 20 mg/kg/d,metformin suspension 300 mg/kg/d gavage,8 weeks after administration to detect the relevant indicators for the detection of materials.Results Compared with the model group,the levels of FBG,HbA1c,TC,TG and FFA were decreased,and the expression of PDX-1 and MaFA mRNA was increased(P<0.05 or P<0.01).Conclusion The mechanism of Dachaihu Decoction in the treatment of type 2 diabetes may be to inhibit oxidative stress and protect the isletβcells.
