金毛狗转录组测序及生物信息学分析 被引量:1

摘要 【目的】利用二代高通量测序技术获得金毛狗的转录组信息。【方法】金毛狗叶片总RNA经过提取、建库、测序和组装,获得单基因簇(Unigene)序列,再进行注释和分析,得到金毛狗转录组信息。【结果】共获得877918条金毛狗Unigene,平均长度为697.85 bp,N50为1262 bp。其中,33682条(38.37%)Unigene在数据库中获得功能注释,并与江南卷柏的序列相似度最高。分别有20379条和25246条Unigene在京都基因与基因组百科全书(KEGG)和基因本体论(GO)数据库获得注释,包括代谢过程、刺激响应、生物调节、转运活性等。此外,4605条1000 bp以上Unigene经过卫星识别工具(MISA)进行简单重复序列(SSR)检测,共鉴定到6456个SSR位点,并使用Primer 3.0软件设计引物。【结论】金毛狗的转录组信息为其生物学过程、次生代谢通路和功能基因的挖掘提供了遗传资源信息,有利于其资源保护和开发利用。 Objective To obtain transcriptome information of Cibotium barometz(L)J.Sm.using second-generation high-throughput sequencing technology.Methods The total RNA from Cibotium barometz(L)J.Sm.was extracted,data-built,sequenced and assembled to obtain unigene sequences,which were then annotated and analyzed to obtain transcriptome information of Cibotium barometz(L)J.Sm..Results A total of 877918 unigenes were obtained,with an average length of 697.85 bp and an N50 of 1262 bp.A total of 33682(38.37%)unigenes were functionally annotated in the database and showed the highest sequence similarity to that of Selaginella moellendorffii.Of these,20379 and 25246 unigenes were annotated in the Kyoto Encyclopendia of Genes and Genomes(KEGG)and Gene Ontology(GO)databases respectively,including metabolic processes,stimulus response,bioregulation,and transport activity.In addition,4605 unigenes over 1000 bp were subjected to simple repeat sequence(SSR)detection by satellite identification tool(MISA),and a total of 6456 SSR sites were identified and primers were designed using Primer 3.0 software.Conclusion The transcriptome information of Cibotium barometz(L)J.Sm.provides genetic resource information for the mining of its biological processes,secondary metabolic pathways and functional genes,which is beneficial to its resource conservation and exploitation.
