摘要
目的:评价利胆排石片中金钱草替以广金钱草或聚花过路黄后对结石模型小鼠结石防治的药效。方法:利用致石饲料连续喂饲8周,制备结石模型小鼠;依照《中国药典》制备利胆排石片(简称LDPS),及广金钱草替代金钱草的利胆排石片(简称LDPS-G)、聚花过路黄替代金钱草的利胆排石片(简称LDPS-J),3个药物组动物造模的同时灌胃上述3种药物,所有动物摘眼球采血,酶标法检测血清中的总胆固醇、钙的浓度,剖取肝胆,观察其形态、胆囊中胆结石的形成。结果:与空白组比较,模型组、LDPS-G组、LDPS-J组动物胆囊体积明显增大,模型组成石率100%,LDPS组成石率20%,LDPS-G组成石率40%,LDPS-J组成石率60%,除LDPS-J组外均可见大型结晶;与空白组比较,模型组小鼠血清中钙离子浓度显著降低(P<0.05)、总胆固醇浓度增加非常显著(P<0.01),3个药物组小鼠血清中钙离子、总胆固醇浓度无明显差异;与模型组比较,其他3个药物组血清中的钙离子浓度均显著升高(P<0.05),LDPS、LDPS-J组血清中总胆固醇浓度降低非常显著(P<0.01),LDPS-G组降低显著(P<0.05);与LDPS组比较,其他两个药物组的钙离子、总胆固醇浓度无明显差异(P>0.05)。结论:LDPS、LDPS-G、LDPS-J对胆结石的形成均有一定抑制作用,LDPS可能通过促进胆汁排出、降低血胆固醇浓度起到防治结石作用,但其抑制结晶的聚集生长作用不明显;LDPS-G抑制结晶的聚集作用不明显;LDPS-J虽然对胆结石形成率较低,但能抑制细小结晶的进一步聚集,防止大型结石形成。
Objective:To study the effect of Lidan Paishi Tablets of Desmodium radiata or Lysimachia congestiflora instead of Lysimachia on prevention and treatment of gallstones in mice.Methods:The Stone feed was fed continuously for 8 weeks,Prepared stone model mice.According to the"Chinese Pharmacopoeia",we prepared three kinds of pharmaceutical preparations.one is Lidan Paishi Tablets,which including Lysimachia(referred to as LDPS),the other is the preparation,which including Desmodium radiata instead of Lysimachia in Lidan Paishi Tablets(referred to as LDPS-G),and the third is the drug,which including Lysimachia congestiflora instead of Lysimachia in Lidan Paishi Tablets(referred to as LDPS-J).While modeling animals,three pharmaceutical preparations were given at the same time.All the mice were bled by eyeball removal.ELISA assay were done to detect the concentration of total cholesterol and calcium in serum.the shape of the liver and the formation of gallstones in the gallbladder were observed.Results:Compared with the blank group,The gallbladder volume of model group,LDPS-G group and LDPS-J group increased significantly;The model composition rate is 100%,LDPS composition rate is 20%,LDPS-G composition rate is 40%,and LDPS-J composition rate is 60%;Large crystals can be seen except for LDPS-J group.Compared with the blank group,The serum calcium concentration in the model group decreased significantly(P<0.05),the total cholesterol concentration increased significantly(P<0.01),and there was no significant difference in serum concentration of calcium ion and total cholesterol in 3 preparation groups.Compared to the model group,Calcium concentrations in the serum of the other three preparation groups increased significantly(P<0.05);The concentration of total cholesterol in serum of LDPS、LDPS-J group decreased significantly(P<0.01),and LDPS-G group decreased significantly(P<0.05).Compared with LDPS group,and there was no significant difference in calcium ion and total cholesterol concentration between the other two drug groups(P>0.05).Conclusion:The LDPS,LDPS-G and LDPS-J all have a certain inhibitory effect on the formation of gallstones.LDPS may play a role in preventing and treating stones by promoting bile excretion and reducing blood cholesterol concentration.LDPS and LDPS-G have no obvious effect on inhibiting the aggrega-tion and growth of lithiasis;LDPS-J has a weaker rate of formation of gallstones,but it can inhibit the further aggregation of fine lithiasis and prevent the formation of large stones.
