措勤紫绒山羊线粒体基因组全序列测定及分析

Determination and Analysis of the Complete Mitochondrial Genome Sequence of Cuoqin Purple Cashmere Goat(Capra hircus)

摘要 措勤紫绒山羊是中国优良的绒用地方山羊品种,从线粒体水平分析其与其他中国地方山羊品种之间的系统发育关系,对措勤紫绒山羊的种群分类和遗传资源保护具有重要意义。本研究利用全基因组随机测序技术和生物信息学软件对措勤紫绒山羊线粒体基因组进行获取、拼接与注释。结果发现措勤紫绒山羊线粒体基因组全长16641 bp,主要由22个tRNA基因、2个rRNA基因、13个蛋白编码基因和1个非编码控制区(D-loop区)4部分构成;基于线粒体基因组D-loop区序列构建8个地方山羊品种的系统进化树,发现措勤紫绒山羊与柴达木绒山羊的遗传距离最近,推测是由地理位置相近所导致的。本研究为措勤紫绒山羊的品种鉴定和系统发育关系提供了一定的数据支撑。 Cuoqin purple cashmere goat is an excellent cashmere local goat breed in China.It is of great significance to analyze the phylogenetic relationship between Cuoqin purple cashmere goat breed and other Chinese local goat breeds from mitochondrial level.In this study,the whole-genome random sequencing technology and bioinformatics software were used to obtain,splice,and annotate the mitochondrial genome of Cuoqin Purple cashmere goat.The results showed that the total length of the mitochondrial genome of Cuoqin purple cashmere goat was 16641 bp,which was mainly composed of 22 tRNA genes,2 rRNA genes,13 protein-coding genes,and 1 non-coding control region(D-loop region).Phylogenetic trees of 8 local goat breeds were constructed based on d-loop sequences of mitochondrial genome.It was found that the genetic distance between Cuoqin purple cashmere goat and Chaidamu cashmere goat was the closest,which was speculated to be caused by the close geographical location.This study provided some data support for variety identification and phylogenetic relationship of Cuoqin purple cashmere goat.
