摘要
目的探讨DJ-1和PTEN在结肠癌组织中的表达及两者的相关性,并探讨其临床意义。方法通过免疫组化法检测DJ-1和PTEN在结肠癌及癌旁组织中的表达及其定位,分析其表达与结肠癌不同临床病理参数的相关性,探讨两种因子之间的表达差异。结果在结肠癌组织及其癌旁组织中DJ-1阳性表达率分别为67.5%和10.0%(P<0.05),PTEN阳性表达率分别为55.0%和95.0%(P<0.05),且DJ-1与PTEN在结肠癌中的表达呈负相关(r=-0.33,P<0.05)。两者的表达与结肠癌的分化程度、淋巴结转移、脉管浸润等密切相关(P<0.05),与年龄、性别、肿瘤部位、大小无关。结论DJ-1在结肠癌组织中的高表达及PTEN在结肠癌组织中的低表达与结肠癌的发生及转移相关,联合检测DJ-1与PTEN对预测结肠癌的恶性程度和浸润转移有重要的临床意义,并且有望成为结肠癌评估预后的生物学指标,并为结肠癌的治疗提供潜在靶点。
Objective To investigate the expression of DJ-1 and PTEN in colon cancer tissues and their correlation.Methods The expression and localization of DJ-1 and PTEN in colon cancer and adjacent tissues were detected by immunohistochemical,and the correlation between the expression of DJ-1 and PTEN and different clinical pathological parameters of colon cancer was analyzed.Results The positive expression rates of DJ-1 in colon cancer tissues and adjacent tissues were 67.5%and 10.0%,respectively(P<0.05).The positive expression rate of PTEN was 55.0%and 95.0%respectively(P<0.05).DJ-1 and PTEN expressed in colon cancer tissue were negatively correlated(r=-0.33,P<0.05).The expression of DJ-1 and PTEN was closely related to the degree of differentiation of colon cancer,lymphnode metastasis and vascular infiltration(P<0.05),but had nothing to do with age,sex or clinical stage.Conclusion The overexpression of DJ-1 and low expression of PTEN are closely correlated with carcinogensis,progressiona and metastasis of colon cancer.Combined detection of DJ-1 and PTEN has important clinical significance in predicting the malignancy and metastasis of colon cancer,and is expected to be a biological marker for evaluating the progrosis of colon cancer,providing a protential target for the treatment of colon cancer.
