强壮灵合剂急性毒性及抗菌抗炎作用研究

摘要 目的探索强壮灵合剂的急性毒性及抗菌抗炎作用。方法采用经典急性毒性实验方法考察强壮灵的急性毒性;通过体内、体外抗菌实验,观察强壮灵的抗菌作用;采用冰醋酸致腹腔毛细血管通透性增加小鼠模型,考察强壮灵的抗炎作用;采用脂多糖所致小鼠脓毒症模型,考察强壮灵对脓毒症小鼠生存率及体内炎症介质的影响。结果强壮灵的最大给药量为每日367 g生药/kg。强壮灵对金葡菌感染小鼠具有明显的保护作用,对0.7%冰醋酸溶液所致毛细血管通透性增加具有明显的抑制作用,可显著提高脂多糖诱导的脓毒症小鼠生存率,明显降低脓毒症小鼠体内MDA、TNF-α、IL-6含量,显著提高SOD活性。结论强壮灵合剂未见明显急性毒性,且可通过抑制体内炎症介质而发挥抗炎、抗细菌感染、抗脓毒症效应。 Objective To observe the acute toxicity,antibacterial and anti-inflammatory effects of Qiangzhuangling Mixture(强壮灵合剂).Methods The classical acute toxicity test method was used to investigate the acute toxicity of Qiangzhuangling Mixture;the anti-bacterial effects of Qiangzhuangling Mixture were observed through in vivo and in vitro anti-bacterial experiments;the anti-inflammatory effects of Qiangzhuangling Mixture were investigated by using acetic acid induced mouse peritoneal capillary permeability model;and the effects of Qiangzhuangling Mixture on the survival rate and inflammatory mediators in mice with sepsis induced by lipopolysaccharides were investigated.Results The maximum dosage of Qiangzhuangling Mixture was 367 g/kg(equivalent to raw drug quantity)per day.Qiangzhuangling Mixture had obvious protective effect on mice infected with Staphylococcus aureus.It had obvious inhibitory effect on capillary permeability caused by 0.7%glacial acetic acid solution.It could significantly improve the survival rate of mice with sepsis induced by lipopolysaccharides,significantly reduce the contents of MDA,TNF-αand IL-6,and significantly increase the activity of SOD.Conclusion Qiangzhuangling Mixture has no obvious acute toxicity,and has obviously anti-inflammatory,anti-bacterial infection and anti-sepsis effects by inhibiting the level of inflammatory mediators in vivo.
