摘要
目的预测中药郁金治疗抑郁症的分子作用机制。方法以网络药理学方法为基础,通过收集中药成分以及抑郁症疾病靶点数据完成靶点预测,进而构建"成分-靶点"网络与"蛋白质-蛋白质相互作用"网络,完成基因功能和通路富集分析。结果筛选发现郁金中含有谷甾醇、β-谷甾醇等4个成分,以上成分共有69个与抑郁症相关的重合靶点,通过David网站富集分析共获得19条具有统计学意义的相关通路。结论中药郁金对于抑郁症具有治疗作用,其中五羟色胺受体、阿片受体等为郁金治疗抑郁症的主要候选靶点,对神经活性配体-受体相互作用、环磷酸腺苷信号通路、钙离子信号通路和多巴胺能神经突触进行网状综合调节为郁金治疗抑郁症的主要调节机制。
Objective To predict the molecular mechanism of turmeric in treating depression.Methods Based on the network pharmacology method,the target prediction was completed by collecting the target data of TCM components and depression diseases,and then the"component-target"network and"protein-protein interaction"network were constructed to complete the gene function and pathway enrichment analysis.Results Four components including sitosterol and beta-sitosterol were found in turpentine.There were 69 overlapping targets related to depression.A total of 19 correlated pathways with statistical significance were obtained through the enrichment analysis of David’s website.Conclusion Turmeric has therapeutic effect on depression.Among them,serotonin receptor and opioid receptor are the main candidate targets of turmeric in the treatment of depression.The network synthesis regulation of neuroactive ligand-receptor interaction,cAMP signaling pathway,calcium signaling pathway and dopaminergic synapse is the main regulatory mechanism of turmeric in the treatment of depression.
