益气消征方对Lewis荷瘤小鼠肿瘤抑制及免疫调节影响的实验研究

Experimental Study on the Effect of Yiqi Xiaozheng Decoction on Tumor Suppression and Immune Regulation in Lewis Tumor-bearing Mice

目的:通过对小鼠Lewis肺癌移植瘤的相关生化指标检测,探讨自拟益气消征方对其肿瘤抑制及免疫功能的调节影响。方法:选取SPF级C57BL/6小鼠,采用动物移植性肿瘤实验法,在实验小鼠右腋下接种Lewis肺癌细胞,从而成功制备小鼠荷瘤实验模型。给予高、中、低不同剂量的自拟益气消征方,分别检测抑瘤率;小鼠胸腺指数、脾指数、肝指数;ELISA法检测血清中肿瘤坏死因子α(Tumor necrosisfactor,TNF-α)、白细胞介素2(Interleukin-2,IL-2)和白细胞介素6(Interleukin-6,IL-6)等细胞因子的含量。结果:①益气消征方和DDP对Lewis肺癌荷瘤小鼠均显示了明显的肿瘤抑制作用,差异有统计学意义(P<0.05),高、中、低剂量中药组组间比较差异部分有统计学意义(P<0.05);②DDP组、高、中剂量组脾指数、胸腺指数、肝脏指数较肿瘤对照组增高,且差异具显著性(P<0.05);③DDP组及益气消征方所有剂量组别血清TNF-α、IL-2和IL-6水平,较肿瘤对照组均明显下降,差异有统计学意义(P<0.05)。结论:自拟益气消征方各剂量组对小鼠Lewis肺癌移植瘤有较好的抑制率,从而起到抗肿瘤的临床效果。

Objective:To investigate the effect of Yiqi Xiaozheng decoction on tumor inhibition and immune function of Lewis lung cancer transplanted tumors in mice.Methods:SPF C57 BL/6 mice were selected and Lewis lung cancer cells were inoculated into the right axilla of the experimental mice by animal transplantation method.The experimental model of mice bearing tumors was successfully established.The anti-tumor rate,thymus index,spleen index and liver index of mice were detected by different doses of self-made Yiqi Xiaozheng decoction,and the contents of TNF-α,IL-2 and IL-6 cytokines in serum were detected by ELISA.Results:①Both Yiqi Xiaozheng decoction and DDP showed significant inhibition on Lewis lung cancer-bearing mice(P<0.05);There were significant differences among high,middle and low dose groups(P<0.05);②Spleen index,thymus index and liver index in high and middle dose groups were higher than those in tumor control group(P<0.05).③Serum levels of TNF-α,IL-2 and IL-6 in all dose groups of Yiqi Xiaozheng decoction were significantly decreased(P<0.05).Conclusion:The self-made Yiqi Xiaozheng decoction has a good inhibition rate on Lewis lung cancer transplanted tumors in mice,and can effectively reduce the angiogenesis rate of tumors.It can protect the cellular immune organs of experimental animalsand enhance the immune function of mice,thus playing an anti-tumor clinical effect.
