逍遥散对慢性应激抑郁大鼠海马一氧化氮合酶表达的影响 被引量:15

Effects of Xiaoyao Powder on the expression of NOS in hippocampus of rats with chronic unpredictable mild stress depression

摘要 目的:探讨逍遥散对慢性不可预知性温和应激(CUMS)抑郁大鼠海马一氧化氮合酶(NOS)表达的影响,以期探讨逍遥散抗抑郁的作用机制。方法:48只雄性SD大鼠随机分为正常组、模型组、氟西汀组和逍遥散组,每组12只。除正常组外,其余各组大鼠每天用CUMS法造模,连续21d,建立CUMS大鼠抑郁模型,在每次造模前30min,给除正常组外的各组大鼠灌胃给药。造模结束后,行糖水偏好实验进行模型评价,并通过Western blot方法从蛋白水平检测各组大鼠海马nNOS、iNOS、eNOS蛋白表达情况,并观测逍遥散对其调节作用。结果:模型组大鼠糖水消耗率显著低于正常组、氟西汀组和逍遥散组(P<0.01),模型组大鼠海马nNOS、iNOS蛋白表达量显著高于正常组(P<0.01),而eNOS蛋白表达量显著低于正常组(P<0.01),逍遥散组和氟西汀组nNOS、iNOS表达均低于模型组(P<0.01),而eNOS表达均高于模型组(P<0.01)。结论:逍遥散能够改善CUMS抑郁模型大鼠的抑郁状态,其机制可能与调节海马内nNOS、iNOS、eNOS蛋白表达量有关。 Objective:To explore the effects of Xiaoyao Powder on the expression of nitric oxide synthase(NOS)system in hippocampus of rats with chronic unpredictable mild stress(CUMS)depression and to explore the mechanism of action of Xiaoyao Powder on depression.Methods:A total of 48 Male SD rats were randomly divided into the normal group,the model group,the fluoxetine group and the Xiaoyao Powder group and with 12 rats in each group.In addition to the normal group,the other groups were established by the CUMS method for 21 days;moreover,rats of all groups except normal group were given gastric lavage 30 min before each model establishment.After the modeling,the model were evaluated by sucrose preference test and Xiaoyao Powder,the protein expressions of hippocampal nNOS,iNOS and eNOS of rats in each group were detected by Western blot,to observe the regulatory effects of Xiaoyao Powder on them.Results:The sucrose water consumption rate of the model group was significantly lower than the normal group,the fluoxetine group and the Xiaoyao Powder group(P<0.01).The expression of nNOS and iNOS in the model group were significantly higher than the normal group(P<0.01),while the eNOS were significantly lower than the normal group(P<0.01),the expression of nNOS and iNOS in the Xiaoyao Powder group and the fluoxetine group were lower than the normal group(P<0.01),while the eNOS were higher than the model group(P<0.01).Conclusion:Xiaoyao Powder can improve the depression status of CUMS rats,and its mechanism may be related to the regulation of nNOS,iNOS and eNOS in hippocampus.
