FAK在胃肠道间质瘤组织中的表达及其与诊断治疗关系的研究 被引量:2

摘要 胃肠间质瘤是胃肠道最常见的间叶源性肿瘤,由于基于肿瘤大小以及核分裂数的提出,单纯的用良性和恶性定义胃肠间质瘤已经没有任何的意义[1]。随着诊断标准的精确化,以往多种消化道肿瘤在相关临床诊断指标中均与GIST相同,应归属于GIST[2]。胃肠道间质瘤的发病和丝氨酸激酶的过度激活有关,免疫组化检测发现GIST过表达CD117和DOG1, 并且大多数病例具有 C-Kit 基因扩增现象[3],KIT 或者PDGFRA基因的持续激活可以启动细胞的过度增殖[4]。然而,在一些KIT或者PDGFRA基因正常的GIST患者中,琥珀酸脱氢酶亚型SDHx(SDHA, SDHB, SDHC, and SDHD)也被证实发生了致病性突变[5]。GIST的发病也和细胞周期的紊乱有关, MDM2或者TP53发生突变,导致细胞周期调节紊乱,这种现象在高危险的胃肠间质瘤中尤其常见[6]。除此以外,一些常见的胃癌致病的环境因素,诸如饮酒、不良饮食习惯亦可能成为胃肠间质瘤的诱导因素[7]。
