摘要
目的 探讨CC族趋化因子巨噬细胞炎症蛋白-1α(MIP-1α)对类风湿关节炎(RA)及骨关节炎(OA)患者体外培养的成纤维样滑膜细胞(FLSs)增殖的影响,以及药物来氟米特(LEF)对FLSs的作用。方法 取体外培养的滑膜细胞第3~5代做实验。流式细胞仪检测FLSs表面MIP-1α受体——CC族趋化因子受体5(CCR5)的表达;MTT法检测加入MIP-1α后FLSs的增殖率;ELISA检测培养FLSs上清液MIP-1α含量;MTT法测定加入药物LEF后FLSs的细胞存活率。结果 RA患者FLSs表面受体表达的阳性率为5.8%~7.6%,OA患者为4.5%~5.6%。RA及OA患者的FLSs在一定浓度的MIP-1α刺激下表现为增殖(P值分别为0.004和0.006)。FLSs培养上清液中有一定含量的MIP-1α,LEF对FLSs的增殖有显著的抑制作用(P<0.05)。结论 RA及OA患者FLSs表面均有MIP-1α受体CCR5的表达。MIP-1α在一定浓度下对FLSs的增殖有促进作用,FLSs可分泌MIP-1α,LEF可显著抑制FLSs增殖。
