摘要
为探讨玉米须黄酮(MSF)简便的提取方法,同时观测其降糖生理活性,本研究采用乙醇浸提法从玉米须中提取黄酮类化合物,对其含量进行测定;在此基础上,以四氧嘧啶致糖尿病小鼠为动物模型,对玉米须黄酮降糖进行动物试验研究。结果表明,醇提法获得玉米须总黄酮的含量为6.81%,该法操作简单方便,无任何化学变化及污染;动物试验揭示玉米须黄酮对四氧嘧啶致糖尿病小鼠具有显著的降血糖和减缓病鼠消瘦症状的作用,这种作用呈现明显的量效关系,效果以高剂量(400mg/kg.bw)最佳。另外,玉米须黄酮能明显提高病鼠超氧化物歧化酶(SOD)活力,减少脂质过氧化物(MDA)的产生,这表明玉米须黄酮的降糖作用机理可能是通过抗氧化作用,减轻四氧嘧啶对胰岛β细胞损伤,或促进已损伤的β细胞的修复,增强胰岛的分泌功能,从而减轻高血糖反应。
A simple extraction method of flavonoids from maize silk(MSF)and its physical activity of controlling blood sugar was studied.The flavonoids was extracted with ethanol and the total flavonoids was determined.The blood sugar control experiment was done on alloxan-induced diabetic mice.The results showed that content of total flavonoids from maize silk was 6.81% and the extraction method was simple and convenient,without any chemical pollution.Animal experiments revealed that flavonoids from maize silk could remarkably decrease the serum glucose in alloxan-induced diabetic mice and was significant dose dependent.Higher dose(400mg/kg·bw treatment)was more efficient.In addition,MSF can improved the activity of serum SOD and decreased their MDA in alloxan-induced diabetic mice.This suggested that the mechanism of MSF may be through anti-oxidation to reduce the injury of alloxan on pancreatic β-cell;it was also possible that the MSF triggered β-cell damage repair system,and enhance the secretion of insulin function to lower the high blood sugar.
