摘要
目的 研究致敏的树突状细胞 (DC)治疗后黑色素瘤 (B16 )小鼠体内TNF -α及IL - 6的变化规律。方法 用冻融抗原致敏BALB/C小鼠骨髓来源的DC。ELISA法检测治疗组荷瘤小鼠、荷瘤未治组小鼠及正常小鼠体内TNF -α及IL - 6水平。结果 随时间荷瘤小鼠体内IL - 6水平逐渐升高 ,经治疗后IL - 6水平升高更为显著 ,而荷瘤未治组小鼠TNF -α水平逐渐降低经治疗后TNF -α水平显著升高。结论 经过抗原致敏的DC治疗 ,荷瘤鼠体内TNF -α、IL - 6水平显著升高 ,表明抗原致敏的DC能够增强荷瘤小鼠抗肿瘤免疫反应。
Objective: To investigate the mechanism in melanoma-bearing mice after treatment by dendritic cells' pulsed with tumor cell lysates.Methods:Dendritic cells generated from murine bone marrow were pulsed with B16 tumor lysates 40 BALB/C mice were divided into 3 groups: normal group without therapy, bearing tumor group without therapy, bearing tumor group with therapy. The level of TNF-α and IL-6 was detected in 3 groups by ELISA. Results: The level of the serum IL-6 of bearing tumor mice was higher than that of control group mice ( P <0.01), furthermore the level of the IL-6 of the mice treated with tumor tysate-pulsed DCs was the highest( P <0.01) in three groups. The level of TNF-α of bearing tumor group was lower than that of control group( P <0.0 1), and the TNF-α level of treated group was obviously higher than that of control group and beating tumor group( P <0.01).The level of IL-6 and TNF-α bearing tumor mice in vivo increased obviously after therapy of tumor lysate-pulsed DCs.Conclusions:Tumor lysate-pulsed DCs enhance anti-tumor immunity effect.
