摘要
目的明确PPARγ天然激动剂姜黄素对自发性高血压大鼠颈动脉中膜组织学变化的影响,分析姜黄素对高血压动脉粥样硬化防治的应用前景。方法 12周龄雄性自发性高血压大鼠20只,随机分为姜黄素干预组(姜黄素200mg/kg.d灌胃给药处理)和模型组(无任何干预的自发性高血压大鼠组),同周龄雄性WKY大鼠10只作为正常血压对照组。大鼠尾动脉无创血压测定方法测定大鼠清醒安静状态下尾动脉的收缩压。喂养至18周龄时终止实验处死动物取颈动脉标本,检测平滑肌肌动蛋白,测定内外弹力膜间血管中膜的厚度、横截面积、平滑肌细胞核面积。结果平滑肌肌动蛋白在3组大鼠中膜中均大量表达;实验开始至实验结束所测的每个时间点均显示模型组大鼠及姜黄素干预组收缩压较WKY大鼠收缩压显著增高,随着鼠龄的增加,模型组及姜黄素干预组大鼠收缩压呈现逐渐增加的趋势,姜黄素干预组和模型组大鼠组在同一时间点收缩压比较无显著性差异;自发性高血压大鼠模型组及姜黄素干预组中膜厚度、中膜横截面积、平滑肌细胞核面积较WKY大鼠增加显著,姜黄素干预组上述3指标较高血压大鼠组明显下降。结论姜黄素作为PPARγ的天然激活剂,能够降低高血压大鼠颈动脉中膜的病理重构,提示姜黄素在动脉粥样硬化的防治中具有一定的应用前景。
Objective To validate the effect of curcumin which is the natural agonist of peroxisome prolifterator-activated receptor(PPARγ) on the carotid media of rats suffering spontaneous hypertension.And to analyze the prospect of curcumin in the prophylaxis and treatment of atherosclerosis.Methods 20 12-week-old male rats suffering spontaneous hypertension were randomly divided to curcumin intervention group(with curcumin 200mg/kg/d irrigation stomach) and model group(rat suffering spontaneous hypertension with no intervention).10 WKY rats with the same age as normal blood pressure control group.Using noninvasive blood pressure measuring method to measure the systolic pressure of rats tail artery when it's awake and quiet.To terminate experiment when rats growed to 18-week old and to execute them to obtain carotid specimens.To detect smooth muscle actin,the thickness of carotid media,cross section area and smooth muscle nucleus'area were measured.Results A lot of expressing of smooth muscle actin in all three groups.From the beginning to the end of the experiment,at each time point,blood pressure of model group and curcumin intervention group was significantly higher than the WKY rats but there was no significant difference between the previous two groups.The systolic pressure of both model group and curcumin intervention group was gradually increased as the age growing.The thickness of carotid media,cross section area and smooth muscle nucleus'area of spontaneous hypertension model group and curcumin intervention group was significantly increased than the WKY rats,and curcumin intervention group was significantly decreased than the spontaneous hypertension model group.Conclusion Curcumin as the natural agonist of PPARγ can reduce pathological reconstruction of carotid media of hypertension rat.It suggests that curcumin have certain application prospect in the prophylaxis and treatment of atherosclerosis.
