摘要
[目的]探讨怀牛膝总皂苷(ABS)大鼠血管平滑肌细胞增殖、迁移的影响,阐明其抗动脉粥样硬化的机制。[方法]采用ox-LDL诱导大鼠主动脉平滑肌细胞增殖的方法,采用MTT法测定VSMC增殖,采用伤口愈合法测定VSMC迁移能力,观察ABS对ox-LDL诱导的大鼠血管平滑肌细胞增殖、迁移的影响。[结果]ox-LDL浓度为30mg/L促进VSMC增殖最强;ABS(50、100、150mg/L)显著抑制ox-LDL诱导的VSMC增殖,作用呈浓度依赖性;ABS(20mg/L)显著抑制ox-LDL诱导的VSMC迁移。[结论]ABS能抑制ox-LDL诱导的VSMC增殖与迁移,说明其具有抑制血管内膜增厚和斑块形成的作用。
[Objective]To investigate the influence of Achyranthes bidentata saponins(ABS)on the proliferation and migration of rats aorta vascular smooth muscle cells,and to elucidate its mechanism of anti-atherosclerosis.[Methods]The oxidized low density lipoprotein (ox-LDL) was used to induce the proliferation of rats aorta vascular smooth muscle cells(VSMC) and Methyl thiazolyl tetrazolium(MTT) assay was used to determine the growth of them,wound healing assay was used to determine the influence of ABS on the ability to migrate of VSMC.All methods were used to observe the effect of ABS on the proliferation and migration of VSMC induced by ox-LDL.[Result]The proliferation of VSMC was at the highest level when the concentration of ox-LDL was 30mg/L,and this effect of ox-LDL could be significantly inhibited by ABS(50,100,150mg/L) in a concentration-dependent manner;ABS could also inhibit the migration of VSMC induced by ox-LDL at 30mg/L.[Conclusion]ABS could inhibit the proliferation and migration of VSMC induced by ox-LDL.From these results,it appears that ABS can reduce the thickness of vascular intima and inhibit the development of vascular plaque.
