莪术对大鼠缺血性脑中风的治疗作用及其机制研究 被引量:7

摘要 目的:探讨莪术对缺血性脑中风的治疗作用及机制。方法:健康成年雄性SD大鼠60只,随机分为假手术对照组、模型对照组、尼莫地平(20 mg.kg-1)组、莪术(16,8,4 g.kg-1)组,每组10只。除去假手术对照组,其余组大鼠采用线栓法制作大鼠大脑中动脉梗阻模型。造模前预防给药7次,造模后给药3次,每天一次。检测大鼠脑含水量、脑梗死体积百分比改变以及脑组织匀浆中MDA、NO的含量和SOD的活力。结果:莪术(16,8,4 g.kg-1)组脑梗死体积百分比和脑含水量为(13.7±1.6)%,(17.4±2.3)%,(24.9±2.9)%和(24.3±4.6)%,(32.2±6.8)%,(33.1±5.6)%,与模型组相比较,莪术(16,8 g.kg-1)组能明显降低大脑中动脉梗阻大鼠的脑梗死体积百分比和脑含水量;MDA、NO的含量为(5.08±0.15),(5.53±0.17),(6.03±0.37)nmol/mgprot和(2.64±0.19),(2.64±0.19),(3.31±0.15)μmol/gprot,与模型组相比较,莪术(16,8 g.kg-1)组能明显降MDA、NO的含量;SOD的活性为(124.14±7.51),(107.07±8.58),(105.42±9.39)U/mgprot,与模型组比较,莪术能明显升高SOD的活性。结论:莪术对大鼠缺血性脑中风有一定的治疗作用,其机制可能与降低脑水肿、抗自由基及保护缺血区脑组织有关。 Objective: To investigate the therapeutic effect and mechanisms of Ezhu on ischemic stroke. Method: Sixty SD rats(maleness) were randomly divided into sham operation control group, model group, nimodipine tablets group, and EZhu(16,8,4 g.kg-1) group.Except the sham operation control group, the middle cerebral artery obstruction model was established in other groups with the suture-occluded method. Each group was treated with corresponding drug 7 times before and 3 times after the model was established.The cerebral water content, the cerebral infarction volume, the content of MDA and NO, the activity of SOD in the brain homogenate were detected. Result:Compared with model group, Ezhu(16, 8, 4 g.kg-1)can significantly decrease the cerebral infarction volume and the cerebral water content to(13.7±1.6)%,(17.4±2.3)%,(24.9±2.9)%,( 24.3±4.6)%,(32.2±6.8)%,(33.1±5.6)% respectively. It can also decrease MDAcontent to(5.08±0.15),(5.53±0.17),(6.03±0.37) nmol/mgprot and NO content to(2.64±0.19),(2.64±0.19),(3.31±0.15) μmol/gprot. But it can significantly improve the activity of SOD to(124.14±7.51),(107.07±8.58),(105.42±9.39) U/mgprot. Conclusion: Ezhu has obvious therapeutic effect on ischemic stroke mouse.The mechanism may be related to reduce cerebral edema, anti-free radical effect and protect the ischemia area of the brain.
