摘要
目的观察龟鹿二仙胶对去卵巢骨质疏松大鼠腰椎骨强度的影响及机制。方法 72只SD大鼠随机分为假手术组(S组)、模型组(OVX组)、雷奈酸锶组(SrR组)、龟鹿二仙胶低剂量组(GL组)、中剂量组(GM组)和高剂量组(GH组),除S组外,其余各组切除卵巢,给药6、12周检测大鼠腰椎骨强度、骨密度、骨小梁面积、平均骨小梁宽度、血清骨钙素、尿脱氧吡啶酚的变化。结果①6周时OVX组腰椎承受最大载荷低于其余各组,12周时GM组明显高于其它2个剂量组(P<0.01)。②12周时龟鹿二仙胶3个剂量组骨密度高于OVX组(P<0.05),低于S组、SrR组(P<0.05);GM组骨小梁面积、平均骨小梁宽度高于其余2个剂量组(P<0.05)。结论龟鹿二仙胶能显著提高去卵巢大鼠腰椎骨强度,以GM组最明显,其作用机制为早期提高骨小梁宽度和骨小梁面积,后期提高骨密度和改善骨显微结构。
Objective To observe the effect of Guilu Erxian glue on lumbar bone strength of ovariectomized rats with osteoporosis and its mechanism. Methods 72 SD rats were randomly divided into sham operation group, model group, Strontium Ranelate group and Guilu Erxian glue groups with different doses. Except sham operation group, other groups received ovariotomy. The changes of lumbar bone strength,bone density, bone trabeculae area, average bone trabeculae width, serum osteocalcin and urinary deoxypyridinoline were detected respectively after 6 and 12 weeks. Results The lumbar strength in model group was lower than that of any other group after 6 weeks, and in middle dosage of Guilu Erxian glue group was higher than that of low and high dosage of Guilu Erxian glue groups after 12 weeks(P<0.01). The bone density in Guilu Erxian glue groups with different doses was higher respectively than that of model group( P<0.05), and lower respectively than that of sham operation group, Strontium Ranelate group(P<0.05). The bone trabeculae area and average bone trabeculae width in middle dosage of Guilu Erxian glue group was higher respectively than that of low and high dosage of Guilu Erxian glue groups after 12 weeks( P<0.05).Conclusion Guilu Erxian glue can enhance the lumbar bone strength of ovariectomized rats significantly, and the effect of middle dosage of Guilu Erxian glue is the best. The mechanism may be related with enhancing width and area of bone trabeculae at the early stage, and enhancing bone density and improving bone microstructure at the later stage.
