摘要
【目的】从11株野生大型真菌中筛选具有产漆酶活性的大型真菌,并对漆酶活性较高的菌株进行产酶发酵条件优化,为将其投入工业化生产提供参考。【方法】以采集自安徽省琅琊山的11株真菌为材料,通过平板显色反应,筛选具有产漆酶活性的大型真菌菌株,再用液体摇瓶发酵培养方法优化大型真菌产酶培养基的组成,并对优化条件进行验证。【结果】1在供试的11株野生大型真菌中,有6株具产漆酶活性,占54.5%;其中有柄树舌灵芝(Ganoderma gibbosum)CZSWXY0001具有较高的产漆酶能力,其产酶培养基的最佳碳源为蔗糖,氮源为酵母膏。2优化后的培养基配方为:蔗糖20g/L,酵母膏2g/L,K2HPO43.2g/L,MgSO4·7H2O 0.2g/L,表面活性剂十二烷基硫酸钠(SDS)3mg/L,Cu2+浓度6mmol/L,pH 7.0。培养基优化后,有柄树舌灵芝菌株所产漆酶活性达到496.18U/mL,约是优化前的12.2倍。【结论】有柄树舌灵芝CZSWXY0001具有较好的产漆酶活性。
【Objective】The specific objectives of the study were to screen laccase-producing wild macrofungi from 11 strains and optimize their optimal production conditions,which could improve the industrialization of laccase-producing strains.【Method】11wild fungi strains collected from Langya Mountain in Anhui were studied to screen strains with laccase-producing ability through the plate-color reaction.Shake flask culturing method was then used to get optimal culture media and conditions.【Result】(1)6out of the11 tested strains had laccase-producing activity with the ratio of 54.5%.Ganoderma gibbosum had the highest laccase-producing ability with the best carbon and nitrogen sources of sucrose and yeast extract,respectively.(2)The optimal medium was:sucrose 20g/L,yeast 2g/L,K2HPO43.2g/L,MgSO4·7H2O0.2g/L,surfactant sodium dodecyl sulfate(SDS)3mg/L,and copper ion concentration 6mmol/L at the pH of 7.0.Enzyme activity under the optimal conditions was 496.18U/mL,which was 12.2times of the original medium.【Conclusion】Ganoderma gibbosum CZSWXY0001 had good-laccase producing ability.
