摘要 以2,2_连氮_双(3_乙基苯并噻唑_6_磺酸)(ABTS)为底物,在pH=4.5的条件下,用分光光度法考察了5种金属离子(Fe2+、Ca2+、Mg2+、Mn2+、Cu2+)对漆酶催化活性的影响。实验发现Fe2+对漆酶的催化活性显示出抑制作用,并进一步探讨了其抑制特征、抑制位点和作用方式。结果表明,Fe2+对漆酶催化活性抑制属竞争性抑制过程,抑制作用是通过还原ABTS·来实现的。 The effect of laccase on ABTS oxidation catalyzed by laccase of five metal ions(Fe 2+ ?Ca 2+ ?Mg 2+ ?Mn 2+ ?Cu 2+ )were studied under the condition of pH4.5.The result showed that the Fe 2+ inhibˉited the catalytic activity of laccase obviously.Its inhibition characteristic,the site and the acting way were studied.The results showed that the nature of inhibition was of a competitive inhibition process.The inhibition was realized through the reduction of ABTS·by Fe 2+ .Other metal ions have small influence on laccase catalytic activity.
