黄芩茎叶总黄酮对小鼠脑缺氧的保护作用(英文) 被引量:10

摘要 背景:研究发现黄芩茎叶总黄酮(TotalflavonoidsfromstemsandleavesofScutellariabaicalensisGeorge,TF)能够增强、改善实验动物学习、记忆能力和降低D-半乳糖产生的神经病理改变,通过腹腔注射NaNO2和断头引起小鼠脑缺氧模型可以进一步验证TF改善记忆能力的作用。目的:研究TF对小鼠脑缺氧的保护作用。设计:完全随机对照实验研究。地点和材料:由承德医学院中药研究所实验动物中心提供的100只昆明小鼠在承德医学院中药研究所生化研究室完成实验。干预:作者用50,100和200mg/kgTF连续灌胃小鼠7d,1次/d,末次给药后60min,各组小鼠分别腹腔注射NaNO2或断头。主要观察指标:以小鼠腹腔注射NaNO2后存活时间和断头后小鼠张口呼吸的持续时间为检测指标。结果:与对照组相比,50,100和200mg/kgTF组小鼠腹腔注射NaNO2后存活时间比盐水对照组分别延长25.3%,50.2%和68.3%;3个剂量组小鼠断头后张口呼吸的持续时间分别是盐水对照组的1.00,1.07和1.48倍,对照组脑复康也显示较强的抗脑缺氧作用。结论:黄芩茎叶总黄酮对小鼠脑缺氧有保护作用。 BACKGROUND:Total flavonoids from stems and leaves of Scutellaria baicalensis George(TF) can enhance and improve learning and memory abilities in experimental animal,and reduce the neuronal pathological alteration induced by D galactose in mice. It is elucidated further the improving effect of TF on memory ability in cerebral hypoxic mice caused by peritoneal injection NaNO2 and decapitation. OBJECTIVE:To study the protective effects of TF on cerebral hypoxia in mice. DESIGN:A completely randomized control trial study. SETTING and MATERIALS: One hundred Kunming mice provided by the Experimental Animal Center,Institute of Chinese Materia Medica,Chengde Medical College were conduced in biochemical laboratory of Institute of Chinese Materia Medica of Chengde Medical College. INTERVENTIONS:Mice were orally administered TF(50,100 and 200 mg/kg,once a day) and Piracetam(PIR,200 mg/kg)for 7 days.The mice were injected NaNO2 and decapitated 60 minutes after last administration,respectively. MAIN OUTCOME MEASURES:The mice's survival time after peritoneal injection NaNO2 and gasping duration after decapitation were as index. RESULTS: Daily oral administration of TF(50,100 and 200 mg/kg) for 7 days could dramatically prolong the survival time after peritoneal injection NaNO2 and the gasping duration after decapitation in mice.Compared with the mice in control group, the survival time after peritoneal injection NaNO2 prolonged 25.3 %,50.2 %and 68.3 %,and the gasping duration was found 1.00,1.07 and 1.48 times,respectively.PIR also revealed the strong anti hypoxic effects. CONCLUSION:TF exerts protective effects on cerebral hypoxia in mice.
