用盾叶薯蓣生产薯蓣皂苷元预发酵与水解条件优化 被引量:38

Optimization of spontaneous fermentation and acid hydrolysis of extracting diosgenin from the rhizome of Dioscorea zingiberensis

摘要  采用RP HPLC法检测薯蓣皂苷元,对薯蓣皂苷元提取过程中影响产率的多种因素,分预发酵、水解2部分分别用单因素和正交设计的方法进行优化。结果表明:40℃预发酵16h,加6%浓H2SO4,料液比为1∶6,温度为121~126℃,水解5h,产率高达3.62%,说明预发酵与水解条件优化可以提高盾叶薯蓣(DioscoreazingiberensisC.H.Wright)生产薯蓣皂苷元的产率。 To optimize the term of extracting technology of diosgenin from the rhizome of Dioscorea zingiberensis C. H. Wright, single factor and orthogonal tests were applied by optimization of extracting technology, the diosgenin was determined by RP-PHLC. The results showed that the optimization terms of spontaneous fermentation is at 40℃, 16 h, acid hydrolysis term 6% H_(2)SO_(4), proportion of solid to liquid 1∶6, temperature 121-126℃, hydrolysis time 5 h. Using these conditions extracting diosgenin from the rhizome of D. zingiberensis, the diosgenin content increased to 3.62%. It suggests the optimization of spontaneous fermentation and acid hydrolysis can improve the production of diosgenin from the rhizome of D. zingiberensis.
