摘要
目的 :观察戊四氮点燃大鼠海马各区巢蛋白 (nestin)的表达 ,以探讨其与癫痫发病机制的关系。方法 :将 2 0只成年雄性SD大鼠随机分为实验组和对照组 ,实验组采用戊四氮点燃大鼠模型 ,观察大鼠痫性发作行为和脑电图表现 ,用免疫组织化学方法观察海马各区星形细胞活化标志巢蛋白表达的变化。结果 :实验组大鼠于 4周后均达到点燃状态 ,而对照组行为正常。 2组脑电图背境电活动以α节律为主 ,实验组Ⅳ或Ⅴ级痫性发作时脑电图均出现典型的高波幅棘慢波、尖慢波。实验组海马各区nestin阳性星形细胞较对照组明显增多。结论 :戊四氮点燃引起海马内nestin阳性星性细胞明显增加 ,提示癫痫脑组织内存在星形细胞增生和活化 。
Aim: To investigate the expression of nestin in different hippocampal areas of pentylenetetrazol kindled rats and explore its relationship with the pathogenesis of epilepsy. Methods: The total of 20 male SD rats were randomly divided into experimental group( n =10) and control group( n =10).The behaviours and EEG expressions were observed.The changes of astrocyte activating marker nestin was examined by immunohistochemical staining in the hippocampus of pentylenetetrazol kindled rats.The examined areas included CA1,CA2,CA3,and the dentate gyrus. Results: The number of nestin positive cells in hippocampus of kindled group were much more greater than those of control group( P <0.05). Conclusion: PTZ kindling induces an increasing in the number of nestin possitive astrocyte,which suggests that proliferation and activation of astrocytes may occur in the epileptic tissue.It may result in scarring and serve as the pathological elements of kindling maintenance.
