红花总黄素对衰老模型小鼠肝线粒体的保护作用 被引量:9

摘要 目的 探讨小鼠肝线粒体膜磷脂组成的随龄变化及红花总黄素 (SY)对肝线粒体膜的保护作用。方法 采用HPLC法测定线粒体膜PC、PE、CL的含量 ,陈思峰法测定PLA2 活性 ,原子吸收法测定线粒体Ca2 +含量。结果 衰老模型组PC、PE、CL均明显降低 ;PLA2 活性随增龄而升高 ;线粒体Ca2 +含量中年组较青年组升高 (P <0 .0 5) ,而衰老模型组Ca2 +含量显著低于中年组 (P <0 .0 5)。SY可升高衰老模型组PC、PE、CL含量和线粒体Ca2 +含量 (P <0 .0 1 ) ;而显著降低衰老模型组PLA2 活性。结论 SY可显著改善衰老模型组肝线粒体膜磷脂的组成 。 Objective To investigate the age related changes of phospholipid contents in hepatic mitochondria membrane of mice and the protective effect of safflower yellow (SY) on hepatocytic mitochondria. Methods We tested the levels of phosphatidyl chorine (PC), phosphatidyl ethanolamine (PE) and cardiolipin (CL) in hepatic mitochondria membrane by high pressure liquid chromatography (HPLC) method, the activity of phospholipase A 2 (PLA 2) by Chen Sifeng's method and the content of Ca 2+ in mitochondria by atomic absorption spectrophotometry. Results The levels of PC, PE and CL of senile model group decreased significantly and the activity of PLA 2increased with ageing. The content of Ca 2+ in mitochondria of middle age group was higher than that of youthful group, lower than that of senile model group ( P <0.05, respectively). SY could enhance the levels of PC, PE and CL and the content of Ca 2+ in mitochondria ( P <0.05) and decrease the activity of PLA 2 of senile model mice significantly.Conclusions SY can obviously improve the phospholipid content in hepatocytic mitochondria membrane, maintain the calcium's homeostasis effectively.
