唐古特青蓝茎愈伤组织的诱导和培养基优化

摘要 目的 进行唐古特青蓝 (DracocephalumtanguticumMaxin)茎愈伤组织的诱导和培养基的优化选择。方法 以唐古特青蓝茎切段为外植体 ,接种在MS和MA二种基本培养基上进行比较实验。在此基础上 ,对 2 ,4 -D、BA、肌醇、CH(水解酪蛋白 )和蔗糖五个因子 ,四种水平进行了正交设计试验。结果 MS培养基诱导唐古特青蓝茎愈伤组织效果较好。其愈伤组织生长的最佳培养基配方为MS +1mg/L 2 ,4 -D +0 .5mg/LBA +5 0 0mg/L肌醇 +6 0 0mg/LCH +3%蔗糖 +0 .7%琼脂。 Objective To study the inducement of Calli from Dracocephalum tanguticum Maxin and the optimization of the Medium. Methods Stalk of Dracocephalum tanguticum Maxin were cut into sections and cultured on MS and MA media, respectively. The optimum composition of the medium for the growth of the calli was screened by using orthogonal design test which included five different factors and four levels. Results The MS medium was better than MA one in terms of inducing calli. The optimum calli growth medium was MS+ 1mg/L 2,4-D+ 0.5mg/L BA + 500mg/L Inositol + 600mg/L CH + 1% Sucrose + 0.7% Agar. Conclusions The optimum medium can improve the growth of calli.
