摘要 目的探讨转化生长因子β1(TGFβ1)II型受体同源序列多肽的生物学功能及其对增生性瘢痕成纤维细胞胶原合成的影响。方法根据TGFβ1II型受体的氨基酸序列人工合成一段短肽,以增生性瘢痕成纤维细胞为靶细胞,分别于蛋白水平和mRNA水平检测该短肽对TGFβ1所诱导的细胞胶原合成的拮抗作用。结果该短肽可以明显抑制TGFβ1所诱导的增生性瘢痕成纤维细胞的胶原合成量及细胞内I型前胶原mRNA的表达。结论该短肽可能作为TGFβ1II型受体的竞争性拮抗剂,抑制或降低了该生长因子对细胞的刺激作用。 AIM: To investigate the biological function of homologous sequence peptide derived from transforming growth factor beta 1 (TGFβ1) receptor type II on the hypertrophic scar fibroblasts.METHODS: This polypeptide was synthesized according to the amino acid sequence of TGFβ1 receptor type II.The antagonistic effect of this polypeptide on TGFβ1 induced collagen synthesis in the fibroblasts from hypertrophic scar was measured acording to both protein and molecule levels. RESULTS: This polypeptide could significantly inhibit the TGFβ1 induced collagen synthesis and down regulate the expression of procollagen I mRNA in the fibroblasts from scar.CONCLUSION: This polypeptide can be taken as competitive antagonist of TGFβ1 receptor II to antagonize or decrease the stimulation of the growth factor to cells.
