摘要
本研究目的是探讨趋化因子受体CXCR4在急性B淋巴细胞白血病 (B ALL)细胞不同分化阶段的表达特点及其与髓系抗原的关系。用流式细胞术采用双色荧光标记法检测B ALL各分化阶段上CXCR4的表达率及平均荧光强度 (MFI)。结果表明 :92 .9%的B ALL表达CXCR4。在B ALL细胞的不同分化阶段 ,CD10、CD34抗原的表达率不同 ,由低到高分化阶段的免疫表型依次为 :①CD10 -/CD34+ ;②CD10 + /CD34+ ;③CD10 + /CD34-;④CD10 -/CD34-。CXCR4在各免疫表型的表达率分别为 (2 7.6 0± 15 .2 5 ) % ,(30 .95± 15 .5 0 ) % ,(5 5 .6 2± 18 37) %和 (77.2 5± 10 .86 ) % ,其MFI分别为 4 6 .6 9± 15 .0 6 ,4 7.4 3± 12 .39,79.2 8± 2 4 .71和 132 .92± 88.0 9。CXCR4在较早期阶段的CD10 -/CD34+ 和CD10 + /CD34+ B ALL细胞上表达无显著性差异 ,均低于CD10 + /CD34-和CD10 -/CD34-的B ALL细胞上的表达 ,在较成熟的CD10 -/CD34-B ALL细胞上表达最高。CXCR4在伴髓系抗原CD13和 (或 )CD33表达的B ALL(my+ B ALL)细胞的表达率为 (12 .5 6± 3.88) % ,MFI为 39.82± 11.5 8,不伴髓系抗原表达组 (my-B ALL)的表达率为 (37.5 7± 11.5 8) % ,MFI为 5 0 .72± 13.34,二者比较有显著性差异。结论 :CXCR4在B ALL细胞上的表达与细胞的?
The study was aimed to explore the chemokine receptor CXCR4 expression on the B-lineage acute lymphocyte leukemia (B-ALL) cells of various differentiation stages and its relationship with myeloid antigen expression. Flow cytometry was used to detect the CXCR4 expression by means of double-flurencence labeling with CD19/SCC gating. The results demonstrated that 92.9% B-ALL patients were positively expressed CXCR4. The CD10,CD34 antigens were differently expressed in differenfiation stages of B-ALL. The immunotypes of (1) CD10 -/CD34 +,(2)CD10 +/CD34 +,(3)CD10 +/CD34 -,(4)CD10 -/CD34 - presented at various differentiatial stages from prematrue to mature. The positive rate of CXCR4 were (27.60±15.25)%,(30.95±15.50)%,(55.62±18.37)% and (77.25±10.86)% from (1) to (4) respectively. The median fluorescence intensity (MFI) of CXCR4 expression were 46.69±15.06,47.43±12.39,79.28±24 71 and 132.92±88.09. CXCR4 expressions were not significantly different between the premature stages of CD10 -/CD34 + and CD10 +/CD34 + subtypes, but both were lower than the CXCR4 expression in CD10 +/CD34 - and CD10 -/CD34 - subtypes. The highest incidence of CXCR4 expression was found in CD10 -/CD34 - B-ALL. The average level of CXCR4 expression on B-ALL cell with positive myeloid antigen CD13 or/and CD33(my +B-ALL) was (12.56±3.88)% of positive rate and 39.82±11.58 of MFI,both of which were less than the positive rate (37.57±11.59)% and the MFI(50.72±13.34) on B-ALL cells with negative myeloid antigen expression(mye -B-ALL). In conclusion,the CXCR4 expression is associated with differentiation level of B-ALL cells and down-regulated when co-expressed with myeloid antigens.
