慢性应激对大鼠外显行为及海马亚区脑源性神经营养因子表达差异性的影响 被引量:15

摘要 目的研究慢性不可预见性应激和孤养结合对大鼠外显行为及海马各亚区结构和BDNF表达的影响。方法应激组经过 2 1d慢性应激后 ,观察所有大鼠行为学改变 ,用ABC法检测海马各亚区的BDNF表达。结果慢性应激后 ,应激组大鼠出现同抑郁症患者相似的行为改变 ;正常组大鼠BDNF在DG区 (光密度为 40 .5 3± 2 .62 ,阳性面积比为 2 .0 13± 0 .0 74)和CA3区 (光密度为 3 9.0 9± 3 .19,阳性面积比为 1.910± 0 .0 5 6)表达明显 ;海马各亚区BDNF同对照组相比明显下降 (P <0 .0 1)。结论慢性不可预见性应激同孤养结合可建立较稳定的抑郁症模型 ;慢性应激可以使海马亚区BDNF表达下降 ;DG区和BDNF在海马可塑性上起到重要作用。 ObjectiveTo study the changes of behaviors , structure and expression of BDNF in hippocampus of rats under the conditions of chronic unpredictable stress and separation. Methods After 21-daystress on the stress group, the behaviors were observed and the levels of BDNF in hippocampus were detected by ABC method for all rats. ResultsAfter chronic stress, depression-like behaviors can be observed and the levels of BDNF decreased significantly in stress group (P<0.01). The high levels of BDNF can be observed in DG zone (optical density was 40.53±2.62,sectional area was 2.013±0.074) and CA3 zone (optical density was 39.09±3.19,sectional area was 1.910±0.056) in control group. ConclusionsThe depressive model can be established steadily by chronic unpredictable stress with separation. Chronic stress can decrease the expression of BDNF in hippocampusof rats. DG zone and BDNF play an important role in hippocampal plasticity.
