急性呼吸综合征患者外周血树突状细胞表型和功能的变化

The alteration of phenotype and function of peripheral dendritic cells in severe acute respiratory syndrome

摘要 目的 探讨严重急性呼吸综合征病毒 (SARSV)感染对树突状细胞 (DC)表型和功能的影响。方法 采用流式细胞仪检测SARS发病期患者 (9例 )和正常人 (7例 )DC的表型 ;分离培养DC和T淋巴细胞 ,用混合淋巴细胞反应检测SARS患者和正常人DC对T淋巴细胞增殖的诱导作用。结果 SARSV感染抑制了DC类刺激分子CD80、CD86和HLAⅡ类分子HLA DR以及与DC成熟有关的分子CD83的表达 ;抑制DC诱导T淋巴细胞增殖的能力。结论 SARSV可抑制DC的分化成熟和抗原递能力。 Objective To explore the pathogenesis of severe acute respiratony syndrome(SARS)Methods The changes of phenotypes of dendritic cells (DCs) were assessed with flow cytometry in 9 SARS patients and 7 healthy persons.The DCs and T-lymphocytes were isolated with gradient centrifugation and cultured in vitro. The proliferation inductions of DCs to T-lymphocytes were measured with mixed lymphocyte reaction.Results Severe acute respiratory syndrome virus suppressed the expression of costimulatory molecules (CD80, CD86), HLA class Ⅱ(HLA-DR) and maturation associated molecule (CD83) on DCs,with significant difference between the two groups,P<0.05. SARS virus inhibited the proliferation function of DC on T-lymphocytes (P<0.05).Conclusion SARS virus can suppress the differentiation and maturation,and the antigen presentation functions of DCs.
