摘要
目的探讨轻度脑损伤早期血清和脑脊液中S-100蛋白(S-100),脑组织中S-100mRNA水平的变化.方法采用雄性SD大鼠应用改良Feeney's方法制造模型,应用聚合酶链反应和酶免疫法动态观察S-100蛋白及其S-100mRNA的变化.结果模型建立后血液中S-100蛋白12,48h分别为(0.825±0.171)μg/L和(1.43±0.082)μg/L,脑脊液中S-100蛋白12,48h分别为(1.21±0.175)μg/L和(1.43±0.082)μg/L,脑组织中S-100mRNA相对灰度值12,48h分别为(25.0±2.6)和(31.4±3.4),与对照组比较都有明显增加(P<0.05).结论 S-100能作为轻度脑损伤的标志物,在损伤后12~48h取样本比较合适.
ObjectiveTo evaluate the level changes of cerebrospinal fluid (CSF), blood S-100 protein (S-100) and S-10mRNA.MethodsMale SD rats were used with Feeney's model by using the RT-PCR technique, the expression of mRNA for S-100 in the brain tissue assay was used to investigate the change of S-100 expression.ResultsThe values of S-100 protein were ((0.825)±(0.171))μg/L and ((1.43)±(0.082))μg/L respectively at 12 hours and 48 hours in the blood, ((1.21)±(0.175))μg/L and ((1.43)±(0.082))μg/L respectively at 12 hours and 48 hours in the CSF after injury. The values of S-100 were increased significantly.ConclusionS-100 is sensitive marker for mild brain injury. The appropriate time for sample collection is 12~48 h after injury.
