摘要
目的 :用TNF α诱导血管内皮细胞 (ECV 30 4 )损伤 ,观察槲皮素对ECV 30 4的保护作用。方法 :采用MTT法观察槲皮素对ECV 30 4活性的影响 :硫代巴比妥酸法测定细胞内脂质过氧化产物丙二醛 (MDA)含量 ;用比色法测定细胞培养液中超氧化物歧化酶 (SOD)和一氧化氮 (NO)含量 ;免疫细胞化学法观察内皮细胞核转录因子 κB(NF κB)的表达。结果 :TNF α对ECV 30 4具有损伤作用 ,槲皮素能显著抑制TNF α诱导ECV 30 4内NO和MDA释放和NF κB的表达 ;并能提高TNF α诱导ECV 30 4内SOD活性。结论 :槲皮素对TNF α诱导ECV 30 4损伤具有保护作用 ,其机制可能与其减少NO释放和抗氧化作用有关 ,而抗氧化作用主要是通过调节NF
Objective: To investigate the effects of quercetin(Que) on the TNF-α injured human umbilical vein endothelial cell line(ECV-304). Methods: The experiments were performed in culture of TNF-α-induced ECV-304 in vitro. Cell viability was assessed with MTT assay. Lipid peroxidation of ECV-304 was monitored as malonaldehyde(MDA) with a fluorometric assay. NO and SOD of ECV-304 were monitored with colorimetry. The expression of NF-κB was detected with immunocytochemistry. Results: TNF-α inhibited the ECV-304 prolifteration. Preincubation of ECV-304 with Que for 24h before TNF-α exposure increased the cell viability. Que significantly decreased the level of NO and MDA and the expression of NF-κB, and increased the activity of SOD in TNF-α-induced ECV-304. Conclusion: These results demonstrate that Que can produce the protective action on TNF-α-induced cultured ECV-304 and its mechanism of action may be related to the decrease of NO, antioxidant effect of lipids. However, antioxidant effect might be exerted through NF-κB activation pathway.
