低温辐射聚合固定化葡萄糖淀粉酶的研究

摘要 研究葡萄糖淀粉酶与α-甲基丙烯酸-β-羟基乙酯单体的低温辐射聚合固定化,观察载体的微孔结构,确定单体浓度(%)在40—60、辐射剂量在5×10~5—1.2×10~6rad、聚合温度在-60—-70℃酶在载体上的担载量在40—70mg/g 之间,所制得的固定化酶的相对活力可达45%以上的最适宜条件,并对工业应用进行了模拟实验. A study is made on the immobilization of glucoamylase and α—hydroxyethyl methacrylate monomer by irradiation polymerization at low temperature.A microhole structure of immobbilized glucoamylase is observed by scanning electron microscopy.The optimal conditions are ascertained as follows:the concentration of monomer ranges from 40% to 60%;the radiation dose ranges from 5×10~5 rad to 1.2×10~6 rad;the temperature of poly-merization ranges from-60℃ to-70℃;and the amount of glucoamylase immobilized on the PHEMA carrier is in the range of 40-70mg/g.With these conditions, the immobilized glucoamylase shows a relative activity over 45%.A simulation test is made for the purpose of industrial application.
