二阶导数分光光度法测定家兔血浆中头孢唑肟浓度及药动学参数

摘要 本文建立二阶导数紫外分光光度法测定家兔血浆中头孢唑肟浓度,并进行药物动力学参数测定。以自制中性氧化铝柱预处理血浆样品,净化回收率86.95±0.66%(n=9),在1~100μg/ml浓度范围内线性良好,r=0.9999,日内日间变异系数低于0.93%,方法回收率为96.05±3.93%(n=9)。家兔耳沿静脉给药(100mg/kg),测得血药浓度经计算机程序处理,符合二室模型,药动学参数分别为:A,36.60±4.94;α,0.115±0.058;B,14.23±6.48;β,0.011±0.005;V(c),2.01±0.31;t1/2,α7.68±4.02;t1/2β,71.68±29.12;K21,0.043±0.028;K10,0.032±0.010;K,12,0.051±0.029;AUC,1672.64±339.17;C1,0.062±0.012。 Determination plasma level of Ceftizoxime in Rabbits by 2 Derivative Spectra photometry was developed in this paper. Plasma samplespreparation was carried out by solidphase cleaned- up through neutral alu-minum column (10×1cm I.D. ) elutingwith distilled water The calibrationcurve was liner (r=0.9999) in the con-cedration range of 1~100μg/ml. Withinday and day-to-day precisions(CV%)were0.91% and 0.93% .The ecroveries obtainedfrom cleaned-up and spiked plasma samplesup to 86.95±0.66% and 96.05±3. 93%,respectively (n=9). There was two-compartment model with i.v. iniection for plasina levels determined throughcomputerized caculation. The pharma-cokinetic parameters were: A, 36.60±4.94; α, 0.115±0.058; B. 14.23±6.48; β,0.01±0.005; V(c), 2.01±0.31; t1/2α,7.68±4.02; t1/2β, 71.68±29.12;K_(21),0.043±0.028; K_(10), 0.032±0.010; K_(12),0.05±0.029; AUC, 1677.64±339. 17;Cl,0.062±0.012.
