反相高效液相色谱法测定山药及其同属植物参薯中薯蓣皂苷元的含量 被引量:19

摘要 目的:建立反相高效液相色谱法测定山药中薯蓣皂苷元含量,考察不同产地山药及其同属植物中薯蓣皂苷元的含量。方法:采用Agilent ODS色谱柱(5μm,4.6mm×250 mm),以甲醇-水(95:5)为流动相,流速1.0 mL·min~1,柱温30℃,检测波长为215 nm。结果:在此色谱条件下,各样品中待测组分均得到良好分离。此法测定的线性范围为5.0—15.0μg,r=0.9999,平均回收率为98.3%。结论:本方法灵敏、准确、回收率高,适用于生药样品检测和质量控制。 Objective: To develop an HPLC method for separation and determination of diosgenin in Dioscorea opposite Thunb.. Methods;The Agilent ODS column(5 μm,4. 6 mm ×250 mm) was used at 30 ℃ with methanol -water(95: 5)as mobile phase. The flow rate was 1. 0 mL · min-1. The detection wavelength was 215 nm. Results: Diosgenin and its primary impurity can be separated obviously. The linearity of the calibration cure was good in the range of 5. 0 - 15. 0 μg for diosgenin and the correlation coefficient was 0. 9999. The average recovery of diosgenin were 98. 3% (n =7). Conclusion:The method is sensitive,reliable,highly reproducible and has been applied to the analysis of Chinese yam grown in different areas with satisfactory results.
