摘要
目的:建立反相高效液相色谱法测定山药中薯蓣皂苷元含量,考察不同产地山药及其同属植物中薯蓣皂苷元的含量。方法:采用Agilent ODS色谱柱(5μm,4.6mm×250 mm),以甲醇-水(95:5)为流动相,流速1.0 mL·min~1,柱温30℃,检测波长为215 nm。结果:在此色谱条件下,各样品中待测组分均得到良好分离。此法测定的线性范围为5.0—15.0μg,r=0.9999,平均回收率为98.3%。结论:本方法灵敏、准确、回收率高,适用于生药样品检测和质量控制。
Objective: To develop an HPLC method for separation and determination of diosgenin in Dioscorea opposite Thunb.. Methods;The Agilent ODS column(5 μm,4. 6 mm ×250 mm) was used at 30 ℃ with methanol -water(95: 5)as mobile phase. The flow rate was 1. 0 mL · min-1. The detection wavelength was 215 nm. Results: Diosgenin and its primary impurity can be separated obviously. The linearity of the calibration cure was good in the range of 5. 0 - 15. 0 μg for diosgenin and the correlation coefficient was 0. 9999. The average recovery of diosgenin were 98. 3% (n =7). Conclusion:The method is sensitive,reliable,highly reproducible and has been applied to the analysis of Chinese yam grown in different areas with satisfactory results.
