摘要
目的:观察穴位电针刺激对面神经再生过程中神经组织神经营养因子-3(NT-3)及其受体表达的影响。方法:日本大耳白兔面神经压榨伤后,电针刺激翳风、颧、地仓、颊车、四白、阳白、合谷穴,每天30min,2周为一疗程。设对照组。经一二三疗程治疗后,应用原位杂交及RT—PCR技术检测针刺组及对照组面神经组织中NT-3及TrkCmRNA表达水平的变化。结果:NT-3、TrkC受体表达高峰均在神经损伤后第6周;在相同时间点,针刺组NT-3、TrkCmRNA的表达明显高于对照组(P<0.05或0.01)。结论:在面神经再生过程中,穴位电针刺激能明显增强面神经组织中NT-3及TrkCmRNA的表达,这一分子水平的表现可能是穴位电针刺激促进面神经再生的机制之一。
Both right and left facial nerve of rabbits were crashed and the degree of the injury was Sunderland IV.Then the injuried rbbits were devided into treatment and the control group.One week after injury,pulse electroacupuncture stimulation on acupoint(PEASA) were performed in teratment group,while the control group was left recovering natruely.the involed acupoints included Yifeng,Quanliao,Jiache,Dicang,Sibai,Yangbai and Hegu.Two weeks to be a course of the treatment.In this experimental modle,we have showed facial nerve regeneration had been improved by acupointpunctrue pathologically and electro physiologically.But on molecular level how about the effects of acupuncture on neurotrophic factors during the nerve regenerate.When the first(3w),second(6w),and third(9w) course of the treatment were finished we used in situ hybridiza tion and RT PCR technique to observe the changes of mRNA expression of NT 3 and Trk C receptors in facial nerve tissue.The results are as follows:(1)mRNA expression of NT 3 and TrkC receptors reaches the highest level in the sixth week post trauma in both treatment and control group;(2)in facial nerve tissue,expression level of mRNA for NT 3 and TrkC in treatment group were signifigantly higher than in the control group ( P <0 01 or 0 05).This experiment result suggests some molecular mechanism about the effects of acupuncture on facial nerve regeneration.
