摘要
目的:建立反相高效液相色谱法测定穿山龙中薯蓣皂苷的含量。方法:采用Hypersil C18(ODS2)色谱柱,乙腈-水(54:46)为流动相,流速1.0 mL·min-1,紫外检测波长为210 nm。结果:薯蓣皂苷可与其他主要成分分离,薯蓣皂苷与峰面积呈良好的线性关系,线性范围为2.3-11.5μg,r=0.9999;样品平均回收率为98.6%,RSD为1.0%(n=5)。结论:本方法用反相高效液相色谱法直接测定了全国6个地区共8个批次的穿山龙中薯蓣皂苷的含量。该方法无需衍生化,分离效果好,分析快速、准确,灵敏度高,重现性好。
Objective: To establish a method using RP - HPLC for determination of the content of dioscin in Di-oscorea nipponica Mak. Methods: The content of dioscin of D. nipponica was determined by reverse phase HPLC at 210 nm on a Hypersil C18 ODS2 column(4. 6 mm ×250 mm,5μm)with acetonitrile -water(54:46)as mobile phase and flow rate of 1. 0 mL·min-1 . Results: The linear range of calibration curve was 2. 3 -11. 5μg (r = 0. 9999), the average recovery was 98. 6% with RSD of 1. 0% (re =5) . Conclusion: The method without derivation is simple, accurate and rapid.
