四逆汤对小鼠全脑缺血-再灌注损伤的保护作用 被引量:9

摘要 目的探讨四逆汤对小鼠全脑缺血-再灌注损伤的影响。 方法 将昆明种小鼠18只随机分成3组,每组6只,分别为对照组、再灌注组、四逆汤组。用结扎双侧颈总动脉和给颈部软组织加压的方法造成全脑缺血,对照组只做手术,不造成缺血,观察90min;再灌注组缺血30min,再灌注60min;四逆汤组手术前用四逆汤0.1ml/g体重,每天灌胃,连续3d。然后处死各组小鼠取大脑皮质,制成匀浆,测大脑皮质乳酸脱氢酶和磷酸肌酸激酶活性。 结果 再灌注组大脑皮质乳酸脱氢酶和磷酸肌酸激酶活性显著低于对照组(P<0.01),四逆汤组显著高于再灌注组(P<0.01)而与对照组比较差异无显著意义(P>0.05)。 结论 四逆汤对小鼠全脑缺血-再灌注损伤有明显保护作用。 Objective To explore the protective effects of SINI decoction (SD) on cerebral is-chemia-reperfusion injury in mice. Methods A global cerebral ischemia-reperfusion injury mice model was produced by blocking bilateral common carotid arteries and pressing cervical soft tissue for 30 minutes, then reopening the vessels and observing for 60minutes. SD was fed at the dosage of 0.1ml/g.wt.d for three days preoperatively. The activity of lactate dehydrogenase (LDH) and creatine phosphokinase (CPK) in cerebral cortex were determined. Results The activity of LDH and CPK in cerebral cortex in reperfusion group were significantly lower than that in control group (P < 0.01), while they were significantly higher in SD group than that in reperfusion group (P < 0.01). No obvious difference was found between SD group and control group (P > 0.05). Conclusion SD is effective in protection of cerebral ischemia-reperfusion injury in mice.
