摘要
建立了茶叶中5种酚酸类化合物的HPLC测定方法,并分析比较了茶叶酚酸类化合物的前处理方法。茶叶中5种酚酸类化合物的HPLC测定条件为:色谱柱采用HypersilBDSC18柱(250×4.6mmID;5μm),流动相为75%A液(2%HAc)+25%B液(乙腈),柱温30℃,流速1.0mL/min,检测波长326nm和280nm,外标法定量。
Five phenoic acids in tea were determined by HPLC, and the effect of the extraction solvent on the sample preparing was studied. Results showed that gallic acid, chlorogenic acid and ellagic acid in tea were higher than caffeic acid and p-coumaric acid. And formic acid -methanol(5∶95,V/V) were the best for sample preparation. But for gallic acid, water is better than formic acid-methanol(5∶95,V/V) as extraction solvent. The samples were analyzed by a Hypersil BDS C18 column (250×4.6mmID; 5μm). The mobile phase was 2% acetic acid:acetonitrile (75:25). The flow rate was 1.0mL/min. Column temperature was 30℃.UV-detection was performed at 326nm and 280nm.
