补肾活血化痰方对AD大鼠学习记忆功能及脑神经元型一氧化氮合酶的影响 被引量:5

摘要 目的 探讨补肾活血化痰方改善学习记忆功能的作用机理。方法 15月龄Wistar大鼠 6 0只 ,随机分成 6组 :正常组、假手术组 (PBS组 )、老年性痴呆 (AD)模型组、脑复康组 ,补肾活血化痰方低、高剂量组。大鼠双侧前脑基底核 (BasalnucleusofMeynert,NBM)注射鹅膏覃氨酸 (Ibotenicacid ,IBO)损毁前脑基底核造痴呆模型。动物经治疗处理2 8d后 ,各组大鼠进行Morris水迷宫行为学试验 ,并检测大鼠海马区神经元型一氧化氮合酶 (nNOS)阳性细胞数。结果 行为学实验显示 :脑复康组、补肾活血化痰方低高剂量组与其它 3组比较有统计学差异 (P <0 0 1) ;模型组nNOS阳性细胞数明显低于正常组和PBS组 (P <0 0 1) ,高剂量组nNOS细胞的表达近似于正常。结论 补肾活血化痰方能抑制痴呆大鼠模型海马区nNOS神经元受损 ,从而达到提高学习记忆水平。 Objective To observe of “Bushen Huoxue Huatan Prescription”(BSHXHTP)on space learning memory.Methods 15-month wistar rats were randomly divided into normal group,PBS group,model group,piracetam treated group,high-dose and low-dose of BSHXHTP group,then induced by injection of basal nucleus of Meynert (BNM)with ibotenic (IBO)to make model,After treating for four wecks,Morris water maze was used assay the ability of space learning memory and assay the expression of neuronal nitric oxide synthas(nNOS) in brain.Results In the behavior experiment,the significant differences were observed among the model,piracetam treated and normal group(P<0.01);BSHXHTP increased the expression of nNOS in hippocampuss region of corter(P<0.01).Conclusions BSHXHTP possesses a certain improvemeut of the space learning memory level though increased the experession of nNOS in hippocampus region of corter.
