宿主骨髓对同种异体皮质骨爬行替代过程中VEGF表达的影响

摘要 目的研究宿主骨髓对VEGF表达的影响,揭示异体皮质骨移植后的愈合机理。方法新西兰大白兔72只,24只制备深冻异体骨,48只移植异体骨术后1、2、4、6、8、12周取材,对比双侧VEGF表达的差异。结果移植后4周,实验侧扩大的哈佛氏管强阳性表达;6~8周,扩大的哈佛氏管内生长活跃的细胞和新的管状结构等强阳性表达;8~12周,吸收骨陷窝内生长活跃的骨细胞强阳性表达。对照侧6周开始出现少量弱阳性表达的扩大哈佛氏管。双侧对比,阳性表达骨细胞计数均差异性显著。结论(1)异体皮质骨内部的血运来源与外周不同;(2)宿主骨髓通过哈佛氏管,对异体皮质骨内部的血运重建和成骨活动中起着重要作用。 Objective:To observe the contrast of vascular endothelial growth factor(VEGF) expression on both sides and explore the effect of host bone marrow on it.Method:Twenty-four New Zealand rabbits were the suppliers of massive allogenic cortical bone stored by deep freezing. Both radius of the other forty-eight New Zealand rabbits were established 1.0cm bone defect and implanted frozen bone,specimens were procured at 1,2,4,6,8 and 12 week after surgery.The results of VEGF expression were observed,the different of both sides was contrasted.Result:On the experimental sides,the expanded Haverisan canal walls were strongly stained by 4 weeks,the strongly postive VEGF expressions of lively osteoclasts, osteoblasts,osteocytes,new formative Haversian-like systems were detected by 6~8 weeks.By 8~12 weeks,the VEGF expressions of numerous osteocytes in the swelled bone lacunaes were strong positive.On the control sides,the expanded Haverisan canal walls were weak stained by 6 weeks.The stained number of osteocytes was very different on both sides.Conclusion:(1)The interior blood supply of allogenic cortical bone is different from the external surface;(2) The host bone marrow takes main effect on the blood and bone regeneration of massive allogenic cortical transplants by the Haversian canal.
